A palladium-catalysed C–H arylation can create a terphenyl scaffold with two distinct chiral axes in a single transformation. In a 2018 report, a Strasbourg team used the stereochemical information in a chiral sulfoxide-containing biaryl precursor to guide formation of both axes. The result is a three-dimensional framework for bidentate ligand design—not, by itself, evidence of broad industrial use.

What makes the molecule unusual?

The scaffold is an ortho-oriented terphenyl: three linked aromatic rings arranged so the structure has a clamshell-like, three-dimensional shape. Its chirality comes from two aryl–aryl bonds whose rotation is sufficiently restricted to preserve distinct spatial arrangements.

This is atropisomerism, a form of axial chirality. In familiar point chirality, the stereogenic feature is often a tetrahedral carbon; here, it is an axis between aryl groups. With two such axes, the scaffold has two stereogenic elements whose configurations contribute to the molecule’s overall stereochemistry.

How does one transformation establish both axes?

The reported route starts from a biaryl precursor containing a chiral sulfoxide. In the palladium-catalysed C–H activation and arylation sequence, the precursor’s sulfur-centered chirality steers the stereochemical outcome: atroposelective functionalization establishes one axis, and atropo-stereoselective arylation forms the second.

The key advance is the coupled outcome: the researchers set two axial elements in one transformation rather than assembling the full stereochemical architecture through separate axis-forming operations. The available account does not establish a complete catalytic mechanism or provide conditions, yields, or numerical selectivity values, so those details should not be inferred from the headline result.

What can the scaffold be used for?

The paper presents the chiral terphenyl as a platform for designing bidentate ligands. It enabled development of S/N-Biax and the diphosphine ligand BiaxPhos. Bidentate ligands bind a metal at two sites; their three-dimensional geometry can shape the environment around that metal and influence the outcome of an asymmetric catalytic reaction.

Chemistry World reported a proof of concept in which a derived BiaxPhos ligand was used in asymmetric hydrogenation. That is evidence of catalytic utility in a specific demonstration, not proof that the ligand works broadly across reactions or is in industrial deployment. Wider possibilities such as materials applications remain prospective rather than established results of this study.

What remains unresolved?

The reported synthesis accessed two of the four possible stereoisomers. The account identifies selective access to both diastereoisomeric families as a future goal, so the method’s stereochemical reach was not yet comprehensive.

The publication record identifies the paper as first published online on 16 February 2018 and included in the 16 April 2018 issue of Angewandte Chemie International Edition, volume 57, issue 17, pages 4668–4672. The abstract describes the stereochemical result, but the available summary does not establish a full substrate scope, numerical yields, enantiomeric or diastereomeric ratios, or exact reaction conditions. The journal record links the article and supporting information for readers seeking experimental detail.
