Cells can migrate through gaps narrower than their nuclei, but they cannot do so without mechanical risk. Cytoskeletal forces squeeze and bend the nucleus; the nuclear lamina and chromatin resist that deformation, while the nuclear envelope keeps nuclear contents separated from the cytoplasm. These defenses are protective rather than absolute. Severe confinement can tear the envelope, rearrange chromatin and produce DNA damage.

Cells also mount an active response. BAF, Lamin C and ESCRT machinery can help seal an envelope rupture, limiting the time that DNA is exposed to the cytoplasm. Sealing the envelope is not the same as repairing every DNA lesion, however, and DNA damage can occur even when no rupture is detected.

Why a narrow gap is a problem for the nucleus

During migration, actin, myosin and other cytoskeletal systems push or pull the cell body. The cell membrane can usually deform substantially, but the nucleus is comparatively stiff and often becomes the largest obstacle. A 2026 review by Patel and colleagues reports tissue interstitial spaces as narrow as 1–10 micrometres and describes typical nuclei as approximately 10–15 micrometres across. Those values provide scale, not universal dimensions: nuclear size and gap width vary with tissue, cell type and experimental method.

When the cell enters a constriction smaller than the undeformed nucleus, the nucleus must elongate, flatten or bend. Forces transmitted through cytoskeletal attachments act on the nuclear surface. The result depends on how sharply the channel narrows, how long the cell remains confined and how rapidly it moves. A gradual, short constriction may be tolerated differently from a long, rigid pore with abrupt corners.

The nucleus is therefore both a mechanical obstacle and a mechanoresponsive organelle. Its shape and internal organization change as force is applied, and those changes can alter transport, gene regulation and migration behavior.

The layered structures that shield the genome

The nuclear envelope separates two chemical environments

The nuclear envelope consists of inner and outer membranes surrounding the genome. In an intact cell it keeps nuclear proteins and chromatin in a controlled compartment and prevents unrestricted contact with cytoplasmic enzymes. Nuclear pore complexes provide regulated transport rather than an open passage.

A rupture creates a temporary breach in that separation. Nuclear and cytoplasmic contents can mix, nuclear proteins can become mislocalized and cytoplasmic factors can reach DNA. The envelope is thus a physical barrier as well as a regulator of nuclear chemistry.

The nuclear lamina bears load

A meshwork called the nuclear lamina lines the inner nuclear membrane. Lamins connect to membrane-associated proteins and to chromatin, distributing stress across the nuclear surface. Lamina composition affects nuclear stiffness and the way a nucleus responds to compression. In the 2016 primary study of confined cancer-cell migration, reducing lamin levels increased rupture incidence in the tested models, while stronger confinement also increased rupture. The observation does not establish one rupture threshold or rate for every cell type.

Lamina proteins are not simply a rigid shell. Their amount, arrangement and biochemical state differ among cancer cells, fibroblasts, immune cells and developing neurons. A cell may change lamina composition during differentiation or in response to its environment, changing its tolerance for squeezing.

Chromatin contributes mechanical resistance

Chromatin is the DNA–protein material inside the nucleus. Its organization and density influence nuclear mechanics and how force is distributed. When a nucleus is compressed, chromatin can shift and, in severe cases, protrude through a damaged envelope. The foundational 2016 study reported chromatin herniation associated with envelope rupture and DNA damage during confined migration.

Chromatin organization can also change without a visible tear. Mechanical deformation may alter contacts between genomic regions, transcriptional activity and the accessibility of repair or replication factors. Such changes are responses to force, not proof that DNA has been physically broken.

What happens as a cell squeezes through

  1. Force is generated in the cell cortex. Actomyosin contraction and traction against surrounding surfaces push the cell body toward the opening.
  2. The cytoskeleton couples force to the nucleus. Linker proteins transmit movement and tension from cytoskeletal filaments to the nuclear envelope and lamina.
  3. The nucleus deforms. It may elongate, flatten or form a leading protrusion to fit the available geometry. Lamina and chromatin resist excessive strain.
  4. The cell either passes or stalls. If the constriction is too narrow or the nucleus too rigid, migration slows. A 2019 review describes delayed cell-cycle progression after passage through small pores, illustrating that successful passage does not mean the cell is unaffected.
  5. Any envelope damage triggers surveillance and repair. If a transient rupture occurs, repair factors are recruited while the cell may continue moving or pause.

This sequence is not identical in every experiment. Some cells repeatedly cross constrictions; others encounter a single severe deformation. Geometry, confinement time, migration speed and cell state all influence the outcome.

How nuclear-envelope rupture occurs

At sufficiently high strain, the inner and outer membranes can tear. The lamina may fail locally, producing a hole through which chromatin herniates. The breach can be brief or prolonged, and microscopy may detect only events above a particular size or duration. Consequently, “no rupture observed” does not prove that the envelope remained perfectly intact.

Rupture incidence rose with both confinement and lamin depletion in the 2016 study’s cancer-cell models. This is evidence for a mechanical relationship in those models, not a general percentage that can be applied to all migrating cells. A dendritic cell, fibroblast, tumor cell and developing neuron may differ in nuclear size, lamina composition, cytoskeletal force and repair capacity.

During a breach, the normal separation between nucleus and cytoplasm is lost. Nuclear proteins can leak out, cytoplasmic proteins can enter and DNA can encounter damaging conditions. The consequences depend on how quickly the hole closes and on the cell’s ability to tolerate the resulting stress.

How cells seal a damaged envelope

Early recruitment: BAF and Lamin C

The 2026 review describes rapid recruitment of barrier-to-autointegration factor (BAF) and Lamin C at rupture sites before the main repair machinery arrives. Their early presence is consistent with a first-response system that helps stabilize exposed membrane and chromatin. The exact timing and contribution of each factor can vary with the experimental model.

ESCRT-III closes the breach

Endosomal sorting complexes required for transport (ESCRT) proteins, including ESCRT-III, participate in membrane remodeling. Studies of migrating cells report ESCRT-III recruitment to nuclear-envelope lesions and restoration of envelope integrity after rupture. ESCRT activity limits the duration of nuclear–cytoplasmic mixing; it does not guarantee that all molecules have returned to their correct compartment.

Envelope repair and DNA repair are separate outcomes. A sealed membrane can still surround DNA that has suffered a strand break, replication problem or chemical lesion. Conversely, a cell may experience DNA stress without a rupture large enough to detect.

Two routes to DNA damage

Damage that follows a rupture

When the envelope is open, cytoplasmic factors can contact chromatin. The 2026 review identifies TREX1, an endoplasmic-reticulum-associated exonuclease, as one reported route by which envelope breach can contribute to DNA damage. Mislocalized nuclear proteins and altered ionic or enzymatic conditions may add to the stress. These mechanisms were documented in particular studies and should not be treated as the only pathway or as an inevitable result of every rupture.

Damage caused by deformation without a detected rupture

Mechanical strain can interfere with DNA replication and the movement of replication forks. Fork stalling can create lesions even when imaging shows no envelope tear. Compression can also change chromatin organization and impede access by replication or repair factors. For that reason, a study must distinguish “DNA damage after migration” from “DNA damage caused by a documented rupture.”

Assay sensitivity matters. A fluorescent marker may reveal a membrane breach but miss a very short event; a DNA-damage marker may appear after the envelope has already resealed. Timing, marker choice and imaging resolution affect the apparent relationship between rupture and damage.

What damage means for the cell

Possible outcomes span a continuum rather than a single fate:

  • Recovery and continued migration: the envelope reseals, damage is limited and the cell resumes movement.
  • Temporary cell-cycle delay: migration through a small pore can be followed by delayed cell-cycle progression, as discussed in a 2019 review.
  • Persistent genome instability: unrepaired lesions or missegregated chromosomes can affect later divisions.
  • Changed gene regulation: altered chromatin organization and nuclear transport can modify signaling and transcription.
  • Loss of viability: extensive or repeated damage can trigger cell death or permanent arrest.

The same mechanical event can therefore be non-lethal in one context and catastrophic in another. A 2026 Nature study reported non-lethal DNA damage in developing neurons, showing that confined-migration damage is not exclusively a cancer-cell phenomenon. Cancer cells, fibroblasts, dendritic cells and neurons have different survival programs and migration roles, so their responses should not be collapsed into one universal outcome.

Comparison factor Questions to ask Why it changes the result
Constriction geometry How narrow, long, rough or sharply angled is the passage? Geometry determines the shape and magnitude of nuclear strain.
Cell type and context Is the cell a cancer cell, fibroblast, dendritic cell or developing neuron? Lamins, chromatin, cytoskeletal forces and repair programs differ.
Lamina and chromatin Which lamins are present, and how is chromatin organized? These properties set nuclear stiffness and stress distribution.
Rupture detection Was a breach directly observed, and what marker or time resolution was used? Small or brief ruptures may be missed.
Damage linkage Was DNA damage shown to follow rupture, or could replication stress explain it? Deformation can damage DNA independently of a visible tear.
Outcome measured Did the study measure envelope repair, DNA lesions, migration, division or survival? These are distinct endpoints; success in one does not establish success in all.

These distinctions also explain why no single rupture frequency belongs in a general description. A rate is meaningful only with the cell type, confinement geometry, lamin state, imaging method and observation period specified.

What the nucleus does beyond resisting force

The nucleus is not passive cargo. Deformation can change nuclear transport, chromatin contacts and signaling pathways. Mechanical inputs may alter which proteins enter the nucleus, how genes are regulated and whether the cell accelerates, pauses or changes direction. Feedback from the nucleus can therefore influence the migration program itself.

This mechanoresponse can be useful: a cell may sense tissue architecture and adapt its speed or route. It can also impose costs. Repeated deformation, delayed replication or persistent mislocalization of nuclear proteins may accumulate stress even when each individual passage appears successful.

How to interpret the evidence responsibly

Most mechanistic observations come from controlled in vitro confinement systems or from specific tissues and cell types. Such systems isolate geometry and force, but they do not reproduce every feature of a living tissue, including extracellular-matrix remodeling, immune signals, blood flow or neighboring cells. Findings should therefore be described as model-specific unless they have been validated across organisms and contexts.

The strongest conclusion supported across the cited work is layered: the nucleus often limits migration through tight spaces; lamina and chromatin provide mechanical protection; severe confinement can rupture the envelope; BAF, Lamin C and ESCRT-III help restore envelope integrity; and DNA damage can arise through rupture-related exposure or through deformation-associated replication stress. The relative contribution of each route remains context dependent.

Key takeaways

  • A cell can pass a gap narrower than its nucleus only by deforming the nucleus.
  • The nuclear envelope, lamina and chromatin distribute and resist mechanical stress, but none makes the genome invulnerable.
  • Confinement and lamin depletion increased rupture in tested cancer-cell models; that result is not a universal threshold or rate.
  • BAF and Lamin C can appear early at rupture sites, and ESCRT-III helps reseal the envelope.
  • Resealing a membrane breach is not equivalent to repairing DNA.
  • Replication-fork stalling and other deformation effects can damage DNA without a detected rupture.
  • Migration, cell-cycle timing, signaling and survival depend on cell type, geometry and the extent of damage.
