Blue light can help contract certain saturated heterocycles by cleaving a carbon–heteroatom bond and then forming a smaller ring. The 2021 method is not a way to remove an atom from any ring: it was developed for specially designed substrates bearing a photoreactive aromatic ketone. The study shows how light can remodel a molecule’s core framework, including in examples derived from pharmaceuticals, peptides, and sugars.
What the reaction changes
A heterocycle is a ring containing at least one atom other than carbon, commonly nitrogen, oxygen, or sulfur. In this method, researchers start with an α-acylated saturated heterocycle: the ring is functionalized with a ketone-containing group positioned to enable the photochemical reaction. After light excites the ketone, a carbon–heteroatom bond can break; subsequent bond formation closes a smaller ring.
That makes the transformation a form of molecular core editing, rather than ordinary functionalization of a group attached to an unchanged ring. The primary paper describes the process as a visible-light-mediated ring contraction and a variant of the Norrish type II reaction. Its account of the method and substrate classes is available in the 2021 paper in Science.

How blue light helps drive ring contraction
In the reaction setup described by UC Berkeley, blue LEDs excite an aromatic ketone attached to the heterocycle. That photochemical step initiates the bond-cleavage sequence. The resulting intermediate can then form a new bond that closes a smaller ring: the reported nitrogen cases use a Mannich-like closure, while oxygen cases use an aldol-like closure. Berkeley’s explanation is in its account of the light-initiated carbon–heteroatom bond cleavage.

The light is not simply a universal trigger. Richmond Sarpong, the study leader, described selecting LEDs at a wavelength that excites the carbonyl group in the starting material but not in the product. In his words, “We very much walk a fine line by choosing blue LEDs that emit at exactly the wavelength to excite the carbonyl in the starting material but not in the product.” That selectivity is part of the reaction design, not a guarantee that an arbitrary blue lamp will reproduce it.

The Berkeley account describes nitrogen, oxygen, and sulfur bond-cleavage examples and reports late-stage transformations involving pharmaceutical derivatives such as rimiterol and mefloquine. These are demonstrations that the reaction can be applied to complex molecular structures; they do not establish a therapeutic benefit, clinical use, or commercial manufacturing process.

Why the method is not a general-purpose ring-shrinking tool
The starting molecule must be designed to support the chemistry, including a photoreactive ketone group. Chemistry World’s coverage identifies the need for a peripheral aromatic ketone as a limitation. A bare heterocycle, or an arbitrary drug molecule that lacks the required arrangement, should not be assumed to contract under blue light.
Wavelength selection also matters: the reaction needs to excite the ketone in the starting material without driving the same photochemistry in the product. Consequently, the published work does not validate a generic blue LED lamp as a substitute for its reaction setup. The paper establishes a synthetic method and examples, not broad success rates, commercial-scale performance, or later validation. The distinction between the demonstrated chemistry and its limits is also noted in Chemistry World’s 2021 coverage.

Why chemists may find the approach useful
Changing a ring framework can be more consequential than modifying a peripheral group: it offers a way to explore a different molecular shape while retaining much of an existing scaffold. This may be useful when a synthesis has already produced a complex molecule and a chemist wants to test a related, smaller-ring structure. Sarpong said the approach “avoids having to go back to the very beginning of a synthesis to build the type of scaffold that can be accessed here.”
Do these 3 things before closing this tab:
1Clear out junk files and repair common Windows errors2Scan for outdated or missing drivers - takes under a minute3Repair Windows errors before they cause bigger problemsThat potential should be kept in perspective. The reported late-stage examples show synthetic flexibility, not that the method is easier, higher-yielding, or superior to other ring contractions in general. Daniele Leonori, an organic chemist at the University of Manchester, characterized it as “a unique photochemical reaction that allows you to completely change the shape of a molecule under very mild conditions.” The evidence supports a notable reaction strategy with specific substrate and wavelength requirements, rather than a plug-and-play tool for modifying any drug.
