A 2021 Chemical Science study shows exactly what “cooperative” means in supramolecular chemistry: two different recognition systems help one another build a structure that neither system forms efficiently on its own. The researchers combined a hydrogen-bonded rosette with a six-zinc-porphyrin nanoring. At micromolar concentrations, the resulting seven-component complex assembled in solution through the interdependence of hydrogen bonding and metal–ligand coordination.

This is a demonstration of molecular self-assembly, not a finished machine, sensor, light-harvesting device or electronic component. The experiments establish how the designed molecules bind and how their interactions reinforce one another.

What the study built

The work, “Cooperative assembly of H-bonded rosettes inside a porphyrin nanoring,” was carried out by Petr Motloch, Pernille S. Bols, Harry L. Anderson and Christopher A. Hunter. It was published online by the Royal Society of Chemistry on 8 December 2020 and appeared in the 2021 issue of Chemical Science, volume 12, pages 1427–1432. The open-access article is available from the Royal Society of Chemistry; the bibliographic record is indexed by PubMed.

“Supramolecular” means that molecules associate through non-covalent forces rather than being joined into one permanent covalent molecule. In this case, the final object is held together by two kinds of reversible recognition:

Part of the assembly Designed components Interaction that holds it together Role in the final complex
Hydrogen-bonded rosette Three pyrimidine derivatives and three barbiturate derivatives Complementary hydrogen bonds Creates a six-membered, hexapyridine ligand arrangement
Porphyrin nanoring A butadiyne-linked ring containing six zinc porphyrins Pyridine–zinc coordination Captures the rosette inside the ring

The overall stoichiometry is 1:3:3: one nanoring, three pyrimidine derivatives and three barbiturate derivatives. Three molecules of each hydrogen-bonding partner form the rosette; pyridine groups attached to that rosette then point toward the zinc centers in the six-porphyrin ring.

Why “cooperative” is the important result

Cooperation here does not mean that the structure is merely complicated. It means that the two recognition processes are mutually supportive. Hydrogen bonds organize the pyrimidine and barbiturate units into the geometry needed for zinc binding. Coordination to the nanoring, in turn, stabilizes and selects that rosette arrangement. The complete assembly therefore benefits from interactions that are weaker or ineffective when considered in isolation.

The authors examined the component systems separately and together. Their spectroscopy showed that the seven-component complex forms under conditions where the isolated interaction systems do not assemble by themselves without the third component. That dependence is the experimental basis for calling the process cooperative rather than simply additive.

How the assembly forms

The construction can be understood as a sequence of mutually reinforcing recognition events rather than as a one-pot reaction that permanently connects all seven pieces.

  1. Hydrogen-bonding partners recognize one another

Pyrimidine and barbiturate derivatives present complementary hydrogen-bond donor and acceptor patterns. Three molecules of each type associate to make a cyclic rosette. The hydrogen bonds are non-covalent, so the rosette remains reversible and can exchange components in solution.

  2. The rosette presents six pyridine ligands

The derivatives are functionalized with pyridine groups. When three pyrimidines and three barbiturates are arranged in the rosette, those pyridines form a hexapyridine ligand set around the outside of the hydrogen-bonded structure. This functionalization is what connects the otherwise separate recognition motifs.

  3. Pyridine ligands bind zinc centers

Each pyridine can coordinate to a zinc ion in a porphyrin unit. The six zinc porphyrins in the butadiyne-linked nanoring provide the complementary metal sites. Coordination places the rosette inside the ring and anchors the two motifs to one another.

  4. The combined complex reinforces the arrangement

Once coordination occurs, the nanoring helps maintain the rosette geometry, while the preorganized rosette supplies the ligand pattern needed for the ring. This reciprocal stabilization allows the seven-component structure to form at concentrations where the separate systems are not detectably assembled in the same way.

What was measured, and what the measurements show

The evidence is spectroscopic. The authors used ultraviolet–visible–near-infrared (UV-vis-NIR) spectroscopy and proton nuclear magnetic resonance (1H NMR) experiments to follow the components and their interactions.

UV-vis-NIR spectroscopy

Porphyrins have strong, characteristic optical absorptions. Changes in those absorptions can indicate that the porphyrin environment has changed, for example when pyridine ligands coordinate to zinc centers or when a guest is held inside the ring. UV-vis-NIR measurements therefore provide a way to monitor formation of the nanoring–rosette complex in solution.

1H NMR spectroscopy

Proton NMR reports on the chemical environments and motions of hydrogen atoms. Hydrogen bonding, coordination and confinement inside the ring can shift or broaden resonances. Comparing spectra of the individual components with mixtures allowed the researchers to determine whether the expected rosette and inclusion complex were present and to analyze their binding behavior.

Together, the two methods support a solution-phase assembly model. They do not constitute an imaging experiment that directly photographs a single complex, nor do they demonstrate operation as a device.

The quantitative result: effective molarity

The authors report a mean effective molarity of about 200 mM for these rosette complexes in chloroform at 298 K. Effective molarity is a way to express the advantage gained when interacting groups are held near one another by a first binding event. Instead of diffusing independently through solution, the groups have a high local concentration and can make the next interaction more readily.

The number must be read narrowly. It applies to the specific rosette complexes studied, in chloroform, at 298 K. It is not a universal value for supramolecular rosettes, porphyrin rings or hydrogen bonds. Changing the molecular structures, solvent, temperature or concentration could change the measured binding behavior.

Why combining the motifs matters

Hydrogen bonding and metal–ligand coordination have different strengths, geometries and environmental sensitivities. A hydrogen-bonded rosette can encode a precise pattern of donors and acceptors, while zinc–pyridine coordination can connect that pattern to a larger metallosupramolecular framework. Using both lets the researchers separate tasks: hydrogen bonds make the rosette, and coordination installs it in the ring.

The design also illustrates preorganization. If six free pyridines and six metal sites had to find one another independently, many competing arrangements would be possible. The rosette gathers the pyridines into a defined geometry before the nanoring engages them. Conversely, the ring supplies a multivalent metal framework that favors the matching hexapyridine arrangement. The result is not simply twelve unrelated contacts; it is a network in which the geometry of one set of contacts improves the probability and stability of the others.

What the experiment does not demonstrate

The study should not be described as a working molecular machine. No mechanical cycle, powered motion or repeated task was demonstrated. The authors also did not establish that this particular complex functions as a light-harvesting antenna, a molecular wire, a conductor, a sensor or a commercial material.

Porphyrin nanorings are relevant to broader photophysical and electronic research, but potential uses of that class of molecule are not results of this assembly experiment. The reported work stops at designing, forming and characterizing the seven-component complex in solution.

How to read the paper’s significance

The significance is methodological as much as structural. The researchers show that a complicated architecture can be assembled by layering recognition rules, provided the components are chemically designed to fit together. The rosette is not an ornamental feature added to a ring; it is the preorganized ligand that makes the second recognition step possible. The ring is not just a container; its zinc sites help select and stabilize the rosette.

This is useful evidence for designing larger supramolecular systems. A chemist seeking a multicomponent assembly can ask whether one interaction creates the geometry needed for another, whether the second interaction feeds back to stabilize the first, and whether the combined system assembles at lower concentration than either subsystem. Those are concrete tests of cooperation, unlike using the word simply to mean that several bonds are present.

Publication timeline and source context

The article first appeared online in 2020 but belongs to the 2021 journal issue. A contemporary report by Annika Friberg in Chemistry World, published 14 December 2020, described the structure as a cooperative seven-component assembly combining hydrogen bonding with metal–ligand coordination. That report is available at Chemistry World. The primary article remains the appropriate source for the stoichiometry, spectroscopy and effective-molarity qualification.

Bottom line for readers

The rosette demonstrates that cooperative interactions can make a multicomponent supramolecular structure accessible when each recognition motif helps organize the other. Three pyrimidines and three barbiturates hydrogen-bond into a hexapyridine rosette; those pyridines coordinate to six zinc porphyrins in a nanoring; and the resulting seven-component complex forms in solution at micromolar concentrations. The approximately 200 mM mean effective molarity, measured for these complexes in chloroform at 298 K, quantifies that local, mutually reinforcing organization. It is a clear demonstration of cooperative self-assembly—not evidence that the molecule is already a functioning device.
