A 2019 synthetic method gives a directing group on 7-azaindole two jobs in sequence. A carbamoyl group attached to N7 first guides metalation and electrophile trapping at C6. It then relocates to N1, where it enables a second directed metalation–electrophile-quench sequence at C2. The result is a controlled route to 2,6-substituted azaindoles without removing and reinstalling the directing group between the two functionalizations.
The “dance” is a precise name for that N7-to-N1 relocation, not a literal motion and not a general term for every rearrangement in heterocyclic chemistry. The work is a molecular-synthesis proof of concept, including late-stage deuteration of one antipsychotic drug scaffold; it is not evidence of a treatment, clinical benefit, manufacturing process or universal drug compatibility.

What chemists mean by a directing-group “dance”
Directed metalation uses a coordinating group to position a strong base and its associated metal next to a selected aromatic C–H bond. Deprotonation then occurs preferentially at that site, creating an organometallic intermediate that can react with an electrophile. The directing group does not merely activate the ring in a broad sense: its attachment point and geometry help determine which carbon is reached.
In the azaindole method, the directing group is a carbamoyl unit. At the first stage it is bonded to the N7 nitrogen of 7-azaindole. That arrangement directs metalation to C6. Under the reported migration conditions, the same carbamoyl group shifts from N7 to N1. The altered attachment changes which C–H bond is positioned for the next metalation. After the shift, the group directs functionalization at C2.
#1 Best Overall
Calling this a “dance” emphasizes relocation followed by renewed control. The group is not discarded after the first substitution, and the chemist does not need a separate install–remove cycle before addressing the second ring position.
The two-stage sequence at a glance
| Stage | Directing-group attachment | Directed site | Chemical operation | Purpose |
|---|---|---|---|---|
| 1 | N7 carbamoyl | C6 | Regioselective metalation, then electrophile quench | Install a substituent at C6 |
| 2 | Carbamoyl group migrates to N1 | Reoriented for C2 metalation | Second directed metalation, then electrophile quench | Reach the 2,6-disubstituted framework |
| Application | Relocated group retained during the sequence | Deuterium can be introduced at a late stage | Late-stage labeling of an antipsychotic scaffold | Demonstrate use on a complex molecule |
The primary paper describes optimization for C2 and C6 metalation separately and iteratively. The publisher’s abstract states that N7-carbamoyl azaindoles undergo regioselective metalation followed by electrophile quench to furnish C6-substituted derivatives. In the presence of catalytic ClCONR2, the carbamoyl group shifts to N1; a second directed sequence then affords 2,6-substituted azaindoles.
Why 7-azaindole is a useful test case
7-Azaindole is a bicyclic heteroaromatic scaffold containing two nitrogens. Its ring positions are not equivalent, so a method that can address C6 and C2 independently is valuable for constructing substitution patterns used in medicinal-chemistry programs. The challenge is selectivity: a metalation reagent may encounter several acidic or coordinating sites, and the first substituent can change the behavior of the remaining ring.

The migration strategy changes the topology of the same group instead. N7 attachment is used for the first regioselective event; N1 attachment is used for the second. That is the synthetic advantage established by this study.
The approach should not be interpreted as a universal solution to azaindole substitution. The reported sources establish the sequence and its demonstration, but they do not supply a complete protocol, a numerical substrate-scope analysis, broad functional-group-tolerance data or a manufacturing-scale assessment.

How the carbamoyl group changes the reaction map
First: N7 attachment directs C6 metalation
The starting azaindole bears a carbamoyl directing group on N7. Directed metalation produces an organometallic intermediate at the C6 position, and an electrophile is then added to convert that intermediate into a C6-substituted product. “Electrophile quench” describes this trapping step: the carbon–metal bond formed during metalation is replaced by a carbon–electrophile bond.
The key point is positional control. The N7 carbamoyl arrangement favors C6 rather than allowing an uncontrolled mixture of ring substitutions. The source material reports the transformation as regioselective; it does not provide a single performance statistic that can be generalized to every substrate.

After C6 functionalization, catalytic ClCONR2 promotes migration of the carbamoyl group from N7 to N1. The available descriptions establish the net relocation, but they do not present a complete mechanistic account that would justify assigning every bond-making and bond-breaking event. It is therefore safest to describe the chemistry by its observed connectivity change and its synthetic consequence.
The migration changes the directing environment of the bicyclic core. Instead of repeating the N7-controlled C6 operation, the chemist now has an N1-carbamoyl azaindole set up for a second directed metalation. The group’s new location is what makes access to the C2 position possible in the reported sequence.
Third: a second metalation–quench reaches C2
The N1-directed substrate undergoes another metalation and electrophile quench. Substitution at C2, combined with the earlier C6 substituent, gives the 2,6-disubstituted azaindole pattern. The two operations can be considered separately during planning: one establishes C6 selectivity, and the migrated group establishes the conditions for C2 selectivity.
What “iterative” means in this paper
Iterative does not mean that the same reaction is simply repeated under identical conditions. The first and second metalations address different carbons because the directing group occupies different nitrogens at each stage. The sequence is better represented as:
- Prepare the N7-carbamoyl 7-azaindole.
- Perform directed metalation at C6 and quench with the chosen electrophile.
- Use catalytic ClCONR2 to relocate the carbamoyl group from N7 to N1.
- Perform the second directed metalation, now set up for C2, and quench with an electrophile.
- Obtain a 2,6-substituted azaindole bearing the substituents installed in the two quench steps.
The published abstract does not provide reagent equivalents, temperatures, reaction times, isolated yields or a universal order of electrophiles. Those details must be taken from the full experimental article and supporting information rather than inferred from the abstract.
Why moving a directing group can be better than replacing it
Directing groups are powerful but can add synthetic overhead. Installation consumes a step, removal consumes another, and each manipulation can expose a sensitive molecule to additional conditions. A mobile group offers a different economy: the chemist changes its attachment point and reuses its coordinating function.
That benefit is most apparent when two nonadjacent positions on one heteroaromatic scaffold must be differentiated. In this case, N7 attachment selects C6 first, while N1 attachment makes C2 accessible later. The group therefore acts as a switch between two regioselectivity programs.
This is not automatically a shorter or safer process in every laboratory. Migration introduces its own reaction step, and the compatibility of a particular substrate with strong metalation conditions still has to be established experimentally. The study demonstrates a strategy, not a blanket replacement for other azaindole-functionalization methods.

What the late-stage deuteration experiment demonstrates
The authors report late-stage deuteration on an antipsychotic drug scaffold. In a deuteration experiment, a selected hydrogen site is replaced with deuterium, the stable isotope of hydrogen. Such labeling can help trace metabolism, distinguish a synthesized compound from an unlabeled analogue or probe a molecule’s behavior in research.
Here, the demonstration matters because the scaffold is more complex than a simple model azaindole. It shows that the directing-group sequence can be applied late in a synthesis to a drug-related molecular framework. The news coverage describes modification of a commercial antipsychotic agent, while the primary study presents the labeling demonstration as part of the method.
The result should be stated narrowly. One antipsychotic scaffold does not establish compatibility with all medicines, all functional groups or production-scale batches. Nor does deuteration itself establish improved efficacy, safety or clinical value. The evidence concerns synthetic access and isotope labeling.
What the 2019 sources actually establish
- The peer-reviewed method paper was first published online on 15 March 2019. PubMed lists the journal publication in volume 58, issue 22, pages 7313–7317, dated 27 May 2019, with DOI 10.1002/anie.201901724.
- The authors are Michael E. Dalziel, Jignesh J. Patel, Meagan K. Kaye, Jennifer L. Cosman, Matthew O. Kitching and Victor Snieckus.
- The publisher abstract reports N7-carbamoyl azaindole metalation, electrophile quench to C6 products, catalytic migration to N1 and a second sequence that affords 2,6-substituted products.
- The study includes optimization for C2 and C6 metalation as separate and iterative operations and reports late-stage deuteration of an antipsychotic drug scaffold.
- Chemistry World’s 14 May 2019 account places the work in the broader history of migrating directing groups and quotes Victor Snieckus calling it the first application of a directing-group “dance” to azaindoles.
Chemistry World also reports Matthew Kitching’s observation of a red-to-green color change associated with the lithiated intermediate and Eva Hevia’s description of the approach as elegant, with controlled regioselectivity. Those observations provide contemporary context; they are not quantitative performance measurements.
How to compare this method with alternatives
The available evidence supports a focused comparison, not a league table of synthetic methods. Three questions are meaningful:
Best Value
| Comparison question | What this sequence does | What is not established |
|---|---|---|
| Which ring carbon is reached? | C6 first, then C2 after migration | A general efficiency ranking against every C2- or C6-functionalization method |
| Where is the directing group? | N7 before migration; N1 afterward | That every azaindole directing group can migrate in the same way |
| What is the use case? | Iterative electrophile installation and a late-stage deuteration demonstration | Broad drug-library validation, scale-up or clinical utility |
Claims about yield, substrate count, safety, cost, reagent availability or industrial throughput require the full paper and supporting information. Neither the publisher’s abstract nor Chemistry World’s coverage supplies those metrics, so they should not be invented.
How to read the significance without overclaiming
The strongest conclusion is methodological: a carbamoyl directing group can be repositioned on 7-azaindole so that two different ring carbons become addressable in sequence. That solves a real regiochemical planning problem and extends a migration concept previously associated with smaller heterocycles to an azaindole scaffold.
The work is not a drug-discovery result in the clinical sense. It does not report a new therapy, patient data, pharmacological benefit or manufacturing process. Its value lies in showing medicinal and synthetic chemists a way to program two functionalization events on one heteroaromatic core, with isotope labeling as a complex-molecule demonstration.
Bottom line for synthetic planning
If a target requires substitution at both C6 and C2 of 7-azaindole, the reported “dance” offers a logical sequence: use N7-carbamoyl control for C6, migrate the group to N1 with catalytic ClCONR2, then use the new orientation for C2. The 2019 study validates that design and demonstrates late-stage deuteration on one antipsychotic scaffold. It should be treated as a defined proof of concept whose scope, scale and functional-group limits must be checked substrate by substrate.
