A lipoxygenase from the edible freshwater cyanobacterium Aphanothece sacrum has been identified as the gateway enzyme for making saclipins, unusual natural oxylipins that absorb ultraviolet light. The enzyme, AsLOX2, oxygenates the fatty acid α-linolenic acid at two positions and produces the key intermediate 9,16-diHPOTE.
The work, published online in the Journal of Natural Products on September 29, 2026, demonstrates a practical laboratory route to saclipin-like products. It is not yet a finished sunscreen ingredient or a complete reconstruction of the cyanobacterium’s natural pathway.
What the study discovered
Researchers from Meijo University searched the genome of Aphanothece sacrum, known in Japan as Suizenji-nori, for lipoxygenase enzymes that could begin saclipin biosynthesis. They expressed two candidates, AsLOX1 and AsLOX2, and tested them with α-linolenic acid (ALA), linoleic acid and arachidonic acid.

AsLOX1 showed no detectable activity under the tested conditions. The decisive feature was AsLOX2’s ability to add oxygen at both carbon 9 and carbon 16 of ALA, creating the hydroperoxide 9,16-diHPOTE. Chromatographic and mass-spectrometric analyses confirmed the identity of that intermediate.

Under optimized conditions reported by Meijo University in 2026, more than 80% of the starting fatty acid was converted to 9,16-diHPOTE within a few minutes. That figure describes conversion to the intermediate, not the overall yield of isolated saclipins.
How the demonstrated production route works
- Gateway oxygenation: AsLOX2 acts on ALA and introduces oxygen at C9 and C16, forming 9,16-diHPOTE.
- Hydroperoxide reduction: Stannous chloride (SnCl2) reduces the hydroperoxide to the corresponding diol, 9,16-diHOTE.
- Oxidation to saclipins: Manganese dioxide (MnO2) oxidizes the diol and produces compounds matching authentic saclipin A and saclipin B.
Saclipin A was formed in substantially greater quantity than saclipin B, at approximately a 9:1 ratio in the reported product mixture. Because the final two transformations were performed chemically, the experiment establishes a hybrid enzymatic-chemical route rather than the complete biological sequence used by A. sacrum.

Why AsLOX2 matters
It identifies the pathway’s entry point
Finding the gateway enzyme gives scientists a defined starting point for studying how an edible cyanobacterium makes UV-absorbing oxylipins. Before this result, the biochemical step connecting a common fatty acid such as ALA to saclipin structures had not been assigned to a specific enzyme.
It shows unusual dual oxygenation
Many lipoxygenases oxygenate a fatty acid at one principal position. AsLOX2’s C9/C16 oxygenation pattern generates the distinctive 9,16-diHPOTE scaffold associated with saclipins. That selectivity is useful both for mapping the natural pathway and for considering enzyme-based production.

It provides a measurable process step
Conversion above 80% in a few minutes under optimized conditions indicates that the gateway reaction can be efficient in a controlled laboratory assay. It does not by itself establish reactor productivity, enzyme lifetime, purification cost or commercial scale.
What remains unknown about natural biosynthesis
The study does not identify the downstream enzymes that operate inside A. sacrum. The researchers used SnCl2 reduction and MnO2 oxidation to reproduce products corresponding to saclipins A and B, but those reagents may not represent the cyanobacterium’s own catalysts.

- Can the pathway be reconstructed biologically in a host organism?
- How stable and productive is AsLOX2 during extended operation?
- Can the approximately 9:1 saclipin A-to-B ratio be controlled or shifted?
- What purification process would be needed for material suitable for testing?
Could saclipins become natural sunscreen ingredients?
Saclipins absorb ultraviolet light, making them candidates for investigation as UV-filter molecules in cosmetics or protective materials. The Meijo University release also cites earlier observations of antioxidative and antiglycative activity and photo- and thermochemical stability. Those observations help explain the interest in saclipins, but they are not equivalent to evidence that a finished product protects human skin.

No consumer safety profile, regulatory authorization, finished-formula efficacy test or manufacturing-economic analysis has been established by this study. A useful UV filter would need to retain performance in a real formulation, remain stable during storage and use, avoid unacceptable skin or environmental effects, and satisfy the rules governing cosmetic or sunscreen ingredients in the target market.

What the result means for biotechnology
The immediate value is a tractable entry point. Researchers can now test AsLOX2 variants, determine why ALA is preferred over the other tested fatty acids, and search for enzymes that perform the missing downstream transformations. A fully biological route could eventually reduce reliance on stoichiometric reagents, but that outcome has not yet been demonstrated.
The work also illustrates why pathway claims need careful qualification. The greater-than-80% figure concerns an early intermediate, while the 9:1 figure concerns the relative amounts of two products in a chemically completed sequence. Neither number is a production titer for a commercial ingredient.

Publication and timing
Uchida, Taura, Waditee-Sirisattha and Kageyama reported the findings in the Journal of Natural Products, published online September 29, 2026, under the title “Lipoxygenase AsLOX2 Catalyzes the Gateway Oxygenation Step in the Biosynthesis of the UV-Absorbing Oxylipin Saclipin in the Edible Cyanobacterium Aphanothece sacrum” (DOI: 10.1021/acs.jnatprod.6c00875). Meijo University issued a public summary on October 14, 2026.
For now, AsLOX2 should be viewed as a validated gateway enzyme and saclipin production as a promising laboratory route. Turning that chemistry into a natural sunscreen or other UV-protective material will require discovery of the remaining biology, process development and independent safety and performance testing.
