Enzymatic oxidation is usually the greener choice for selectively oxidizing galactose at carbon 6, but it is not automatically the most scalable. Galactose oxidase uses molecular oxygen in water and avoids many hazardous, stoichiometric oxidants. Its industrial prospects depend on oxygen transfer, hydrogen-peroxide control, enzyme lifetime, recovery and downstream purification. Chemical options can be easier to intensify and may work well for bulk galactonic acid, yet supported-gold systems require alkaline operation and precious metal, while electrochemical systems require electrodes, electrolyte and careful byproduct control.
The correct winner therefore depends on the product, reactor boundary and scale. A C-6 aldehyde made with high regioselectivity favors an enzyme. A continuous process for an aldonic acid may favor a recoverable heterogeneous or electrochemical catalyst if its lifetime, salt burden and energy use are acceptable. No cited publication provides a head-to-head life-cycle assessment or plant-scale techno-economic comparison, so claims that one route is universally greener or cheaper would be premature.
What “galactose oxidation” can mean
The comparison is often confused because the two routes may target different products. Galactose contains a primary alcohol at carbon 6 and several secondary alcohols. Oxidizing that primary alcohol gives a C-6 aldehyde. Further oxidation can give galactonic acid, an aldonic acid. A process may also combine oxidation with a later reduction step to reach a rare sugar.

Thus, a reported yield for galactonic acid cannot be compared directly with a yield for a C-6 aldehyde. State the product and oxidation state before comparing methods.
How the enzymatic route works
Galactose oxidase and its reaction
Galactose oxidase (EC 1.1.3.9) is a single copper metalloenzyme with a molecular mass of 65–68 kDa, as described by Parikka, Master and Tenkanen in their 2015 review. It selectively oxidizes the primary alcohol at C-6 of galactose to a carbonyl compound while reducing molecular oxygen to hydrogen peroxide. The enzyme’s value is its combination of aqueous operation and strict regioselectivity: the catalyst recognizes the substrate rather than indiscriminately oxidizing several hydroxyl groups.
Molecular oxygen is a relatively benign terminal oxidant, but it is not a free process benefit. Oxygen must be transferred from the gas phase into the liquid at a rate that matches enzyme demand. The reaction also generates hydrogen peroxide. Accumulated peroxide can inactivate protein, damage product or promote unwanted secondary chemistry, so peroxide removal, decomposition or controlled feeding belongs in the process design from the beginning.
Oxidation–reduction sequences
Some rare-sugar processes use enzymatic oxidation followed by reduction rather than isolating a single oxidation product. A Chemical Reviews article from 2021 reports one such enzymatic system with an 85% final yield and productivity of 0.99 g/L h. Those figures describe that reported system, not a universal performance level for galactose oxidase.
The same review notes that pyranose oxidase can be slow on D-galactose and produce significant byproducts. Pyranose dehydrogenase showed a catalytic efficiency of 46.2 mM−1 s−1 toward D-galactose in the cited comparison—approximately 140 times the reported pyranose-oxidase value. Enzyme choice therefore matters: “enzymatic” is a process category, not one fixed catalyst with one fixed rate.
What makes the enzyme attractive
- Regioselectivity: C-6 oxidation can reduce mixtures of positional isomers and simplify purification.
- Reagent profile: oxygen replaces many harsh, stoichiometric chemical oxidants.
- Operating medium: aqueous, moderate-condition processing is generally compatible with biological feedstocks.
- Engineering options: recombinant expression, protein engineering and immobilization can improve activity, stability, specificity, recovery and reuse.
These advantages do not remove the need for pH control, temperature control, oxygen transfer and peroxide management. DTU process research identifies enzyme stability and oxygen requirements as central barriers to industrial implementation, even though galactose oxidase activity and stability are encouraging.
How the chemical routes work
Supported gold oxidation
Supported gold provides an aerobic heterogeneous route. In the study by Kusema and colleagues, D-galactose was oxidized over Au/Al2O3 in a pH-controlled semi-batch reactor at atmospheric pressure. A mean gold particle size of 2.6 nm gave the highest activity in that study.

Alkaline conditions produced high activity and selectivity toward aldonic acid, whereas acidic conditions inhibited the catalyst and lowered both conversion and selectivity. Alkalinity can improve reaction performance, but it creates a process burden: the plant must handle caustic reagent, neutralization and the salts formed when the product is recovered as an acid or a different salt form.

Gold catalysis is heterogeneous, so filtration, settling or another solid–liquid separation can allow catalyst recovery. That potential advantage depends on support durability, resistance to fouling and retention of nanoscale gold. Precious-metal inventory, losses during operation and end-of-life recovery must be counted in a serious environmental or economic assessment.
Electrochemical oxidation
Electrochemical oxidation supplies the driving force at an electrode rather than adding a separate molecular oxidant. A review summarized a 25-hour, room-temperature electrolysis of D-galactose in 0.1 M NaOH. The reported galactonic-acid yields were 34% on platinum, 67% on a platinum–lead electrode and 86% on gold.
| Electrode | Reported galactonic-acid yield | Conditions and qualification |
|---|---|---|
| Au | 86% | 25-hour room-temperature electrolysis in 0.1 M NaOH; value from the summarized study |
| Pt–Pb | 67% | Same reported electrolysis conditions |
| Pt | 34% | Same reported electrolysis conditions |
Galactaric acid and smaller organic acids were also detected. The results demonstrate that conversion alone is not enough: over-oxidation and electrode selectivity affect the usable product yield. Electrochemical scale-up additionally requires power electronics, electrode area, current distribution, electrolyte circulation, gas management and a plan for electrode replacement or metal recovery.
How broad gold-selectivity claims should be read
A 2017 Chemical Reviews article reports more than 99.5% selectivity to aldonic acids for several cited gold-catalyzed hexose oxidations. That is a useful indication of gold’s potential, not a guarantee for every D-galactose reactor. Support chemistry, particle size, pH, oxygen transfer, substrate concentration, residence time and temperature can all change selectivity. The 99.5% figure must not be transferred to a different reactor without verifying the same product and conditions.

Which route is greener?
Why the enzyme has the stronger inherent green case
For a C-6 aldehyde target, the enzyme starts with several environmental advantages. It uses a selective catalyst in water and molecular oxygen rather than requiring a large stoichiometric quantity of a hazardous oxidant. Fewer positional byproducts can reduce solvent use, reprocessing and waste treatment. The 2021 Chemical Reviews discussion explicitly characterizes an enzymatic oxidation–reduction approach as greener and less hazardous than the corresponding chemical process.
That statement describes the reaction chemistry, not a complete life cycle. Enzyme manufacture consumes feedstock, energy and purification resources. A low-activity enzyme may require a large loading or long residence time. Aeration and agitation consume electricity, and peroxide-control equipment adds materials and operating steps. If the enzyme is discarded after one batch, its environmental advantage can shrink substantially.
When a chemical route can compete
A supported catalyst that runs continuously for a long period, retains its activity and is efficiently recovered may outperform a disposable enzyme on productivity and catalyst utilization. An electrochemical cell can avoid a separate chemical oxidant and can be powered by low-carbon electricity. Those benefits are conditional: alkaline neutralization, salt generation, electrode manufacture, precious-metal demand and over-oxidation must be included in the boundary.

Can galactose oxidase be scaled to industrial production?
Yes, but scale-up is an engineering problem rather than a simple volume increase. A large vessel can have the same chemistry and still fail because oxygen cannot reach the liquid fast enough, peroxide accumulates or the enzyme loses activity before the batch ends.
1. Design oxygen transfer around demand
Measure the oxygen uptake rate at the intended substrate concentration and enzyme loading, then size the gas distributor, agitation and pressure accordingly. Confirm that the volumetric oxygen-transfer coefficient remains adequate at production viscosity and solids content. Air is cheaper than oxygen enrichment, but enrichment or pressurization may be justified if gas–liquid transfer limits productivity. The energy used for compression and agitation belongs in the green calculation.
2. Control hydrogen peroxide
Track peroxide concentration rather than assuming it is harmless because oxygen is the only added oxidant. Options include controlled oxygen feeding, staged substrate addition, in situ peroxide decomposition or a compatible peroxide-scavenging enzyme. Any scavenger must not consume the desired aldehyde, damage the catalyst or complicate purification. Establish a maximum peroxide exposure for both the enzyme and product.

3. Choose soluble or immobilized catalyst operation
Soluble enzyme is simple to mix but can be difficult to recover from a dilute product stream. Immobilization can permit filtration, packed-bed or continuous operation and may improve thermal and operational stability. It can also introduce diffusion resistance, support cost and activity loss during attachment. Compare productivity per reactor volume and per gram of enzyme, not only initial batch conversion.
4. Build a reuse test before committing to continuous processing
Run repeated cycles under realistic feed composition, pH excursions and cleaning conditions. Record residual activity, copper retention, leaching from the support and product carryover. A catalyst that retains 90% activity for two short laboratory cycles may not be economical if it fails after a few full residence times. Conversely, a slower enzyme may be attractive if it remains active for many cycles and avoids expensive downstream separation.
5. Separate product without destroying the advantage
High regioselectivity helps, but the broth still contains protein, salts, unreacted sugar, peroxide-control reagents and potentially oxidation byproducts. Select membrane filtration, adsorption, crystallization or chromatography only after measuring product concentration and impurity profiles. A very dilute, clean reaction can have a worse overall footprint than a more concentrated process that requires a tougher catalyst.
What chemical scale-up requires
Supported gold
Start with a catalyst that reproduces the reported particle-size and support characteristics, then test its stability at the target pH and substrate concentration. The 2.6 nm mean gold particle size was the most active in the cited Au/Al2O3 study, but particle growth, leaching and fouling can change performance over time.

Use a pH-control strategy that accounts for acid formation. If the product is galactonic acid, continuous base addition can hold the reaction in its active alkaline range while generating additional salt. Measure conversion, acid selectivity and carbon balance together; a high disappearance of galactose can simply mean more over-oxidation. Plan catalyst filtration and gold recovery before pilot operation, not after a loss is observed.

Electrochemical cells
Translate laboratory current density and electrode geometry into the electrode area required for the planned production rate. Check whether mass transfer, not intrinsic electrode kinetics, limits conversion. Uniform current distribution becomes harder in large cells, and local hot spots can increase over-oxidation. The reported 25-hour experiment provides useful comparative yields but does not establish long-term electrode life, industrial current density or total energy per kilogram.
Electrolyte selection affects conductivity, corrosion, separation and waste. The cited study used 0.1 M NaOH, so its yields should not be presented as neutral-pH performance. Account for the energy source, rectifier efficiency, pumping, cooling and treatment of alkaline product liquor.
Yield, selectivity and cost: how to compare fairly
Use a common basis such as kilograms of isolated galactonic acid or C-6 aldehyde per kilogram of galactose fed. Report conversion, selectivity, isolated yield, space–time yield and catalyst consumption separately. A high conversion with substantial galactaric acid and small organic acids may produce less saleable product than a lower-conversion process with clean selectivity.

Choose an enzymatic route first when:
- The required product is the C-6 aldehyde and positional purity is more valuable than maximum throughput.
- The feed is sensitive to harsh reagents or the plant wants to minimize hazardous oxidant storage.
- Aqueous processing and moderate conditions reduce corrosion and safety complexity.
- Immobilization or membrane retention can deliver enough catalyst reuse.
- Oxygen transfer and peroxide control can be demonstrated at the intended concentration.
Investigate supported gold when:
- The target is galactonic acid or another bulk aldonic acid.
- A continuous heterogeneous reactor and reliable solid–liquid separation are available.
- Alkaline operation, neutralization and gold recovery are acceptable within the product specification and waste plan.
- Long-term tests show that particle size, support and activity remain stable.
Investigate electrochemistry when:
- Low-temperature operation and direct electrical control are valuable.
- The site has low-carbon electricity and the electrode area can be scaled economically.
- Electrode life, current efficiency and byproduct control are established at production-relevant conditions.
- Alkaline electrolyte and salt handling do not overwhelm the separation train.
A practical development sequence
- Lock the product specification. Define aldehyde versus galactonic acid, acceptable galactaric acid, residual galactose, color, salts and metal or protein limits.
- Run matched carbon balances. Analyze starting sugar, desired product, over-oxidized acids and unidentified organic carbon for every catalyst.
- Measure kinetics under process conditions. Vary substrate concentration, pH, temperature, oxygen availability, enzyme loading or current density rather than relying on one favorable laboratory point.
- Test catalyst lifetime. Use repeated enzyme cycles, supported-gold recycles or extended electrolysis, and quantify activity loss and material leaching.
- Design separation with the reaction. Include protein removal, catalyst filtration, alkaline neutralization, peroxide treatment and salt disposal in the flowsheet.
- Calculate product-based metrics. Compare energy, water, reagents, catalyst make-up and waste per kilogram of isolated product.
- Pilot the limiting operation. For enzymes, that is usually oxygen transfer and peroxide control; for chemicals, it is often catalyst stability, pH management, electrode area or separation.
Common failure modes and fixes
High enzyme activity in a flask but low productivity in the reactor
Likely causes are oxygen limitation, poor mixing, excessive viscosity or peroxide accumulation. Measure dissolved oxygen and peroxide profiles, increase gas–liquid transfer, lower instantaneous substrate loading or use staged feeding. Do not simply add more enzyme until the transfer limit is resolved.
Good galactose conversion but poor aldehyde recovery
Long residence time or peroxide exposure may be driving secondary oxidation. Shorten the reaction, remove product continuously, lower peroxide concentration and verify that any scavenger is chemically compatible with the aldehyde.
Gold catalyst loses selectivity
Check pH drift, particle growth, support fouling and local oxygen or substrate gradients. Re-establish the catalyst’s particle-size distribution and compare fresh and recycled material. A nominally identical gold loading can behave differently if dispersion has changed.

Frequently asked questions
Frequently Asked Questions
Is enzymatic galactose oxidation always greener than chemical oxidation?
No. It has the stronger inherent green profile for selective C-6 oxidation, but enzyme manufacture, oxygen-transfer energy, peroxide treatment, catalyst reuse and purification can change the result. A valid comparison must use the same product and include the full process boundary.
What is the best catalyst for converting galactose to galactonic acid?
The cited electrochemical comparison gave the highest reported galactonic-acid yield on gold at 86%, versus 67% on Pt–Pb and 34% on Pt under 25-hour room-temperature electrolysis in 0.1 M NaOH. Supported Au/Al2O3 also performs well under alkaline aerobic conditions. These results do not establish one universal best catalyst without matching reactor, pH, lifetime and separation requirements.
Does galactose oxidase produce hydrogen peroxide?
Yes. Galactose oxidase uses molecular oxygen and reduces it to hydrogen peroxide while oxidizing the C-6 primary alcohol. Peroxide concentration must be monitored and controlled because it can deactivate the enzyme or promote unwanted chemistry.
Can an immobilized enzyme run continuously?
Immobilization can improve recovery and stability and may enable packed-bed or membrane-retained operation. Diffusion resistance, support cost, activity loss during attachment and cleaning tolerance must be measured before continuous operation is selected.
Why do alkaline conditions help chemical galactose oxidation?
In the cited Au/Al2O3 study, alkaline conditions supported high activity and aldonic-acid selectivity, while acidic conditions inhibited conversion. Alkalinity also creates caustic, neutralization and salt-handling obligations.
Are the reported yields directly comparable?
No. The enzymatic 85% figure is a final yield for one oxidation–reduction system, while the 86%, 67% and 34% electrochemical values are galactonic-acid yields from a 25-hour study in 0.1 M NaOH. Different products, reactors, times and analytical boundaries require caution.
