Microneedles are designed to cross the skin’s outer barrier; nanoneedles and microneedle-based nanomedicine remain research concepts whose readiness depends on the specific product and evidence. They are not interchangeable with ordinary injection needles, and “nanorods” should not be read as a routine drug-delivery treatment.
Why look beyond conventional needles?
A hypodermic needle delivers medicine through the skin into the intended tissue. It is an established injection route, but not every medicine is given by injection. For medicines applied to intact skin, the outer skin barrier limits how readily many substances can pass through. Microneedles are being studied as a way to cross or disrupt that barrier, while nanoneedles and nanomedicine explore different scales and delivery strategies.
The key distinction is not simply needle size. It is where the system is intended to deliver a payload, how it releases that payload, and what stage of evidence supports its use.
How do microneedles deliver medicine?
Microneedles are micron-scale projections designed to cross the skin’s outer barrier, often without reaching deeper structures. Their physical design determines whether they create pathways, carry medicine on their surface, dissolve, or channel a liquid payload. Reviews describe several different approaches, each with its own payload and application constraints (2018 review of transdermal microneedles).

Solid microneedles
Solid projections can create small pathways through the outer barrier. A medicine may then be applied to the treated area or delivered by a separate formulation. The needles themselves do not necessarily contain the dose, so the method involves more than simply placing a drug-coated patch on the skin.
Coated microneedles
In a coated design, medicine is carried on the needle surfaces and transferred during application. The amount that can be carried and released is tied to the coating and device design.

Dissolving microneedles
Dissolving projections contain medicine in a material intended to dissolve after insertion, releasing the payload. This design is being investigated for different payloads, including nucleic-acid therapeutics. Results from preclinical studies do not establish how well a treatment will work in people.
Hollow microneedles
Hollow projections provide a channel through which a liquid formulation can be delivered. Their use depends on the device, formulation, and intended delivery site; they are not simply smaller versions of every hypodermic injection.

What does “microneedle-mediated nanomedicine” mean?
It combines two parts: a microneedle platform that crosses or disrupts the skin barrier, and a nanoscale formulation intended to carry or present the therapeutic or diagnostic payload. The device and the formulation each contribute to delivery. The combination may be studied to address limitations of a formulation or route, but it also raises product-specific questions about material properties, release, safety, and manufacturing. A review describes this as a developing area rather than a blanket clinical capability (2024 review of microneedle-mediated nanomedicine).
Are nanoneedles or nanorods established treatments?
Nanoneedles are an emerging research area. Reviews describe investigations into navigating biological membranes and delivering material inside cells, which is distinct from a conventional injection into tissue (2023 review of nanoneedles in drug delivery). The term “nanorod” is not evidence that a particular structure is a needle-like delivery device or an approved therapy. The available sources support discussion of nanoneedles and microneedle–nanomedicine research, not a claim that nanorods are an established drug-delivery treatment.

For any of these approaches, a research concept, laboratory demonstration, clinical-trial product, and approved medicine are different evidence stages. A review of microneedle-enabled therapeutics and biosensing in clinical trials discusses clinical-trial activity, but that does not establish approval or routine availability for every design (2023 review of microneedle-enabled approaches in clinical trials).
What does the evidence say about dissolving microneedles?
A 2025 systematic review reported that 83% of the included preclinical studies of dissolving microneedles for nucleic-acid delivery found efficacy equivalent or superior to comparator administration routes (2025 systematic review). That figure is the share of studies in a preclinical evidence synthesis. It is not a patient response rate, a clinical success percentage, or a general estimate of microneedle effectiveness.

How does FDA assess medicines containing nanomaterials?
In the United States, FDA’s April 2022 guidance addresses human drug products, including biological products, that contain nanomaterials in the finished dosage form. It discusses characterization, controls, testing, and qualification, and notes that nanomaterials may be active ingredients or inactive carriers with attributes warranting particular examination (FDA, April 2022 guidance). The presence of a nanomaterial alone does not establish a product’s safety, effectiveness, or approval.
FDA’s June 2014 guidance discusses dimensional and dimension-dependent properties as considerations in deciding whether an FDA-regulated product involves nanotechnology (FDA, June 2014 guidance). These are guidance documents, not approvals of a product or a universal classification rule for every microneedle.
How should you compare these delivery approaches?

There is no universal “best” needle type. A meaningful comparison needs the intended tissue and route, payload and formulation, physical design and release mechanism, material and safety considerations, evidence stage, and regulatory status for the specific product and geography.

What should you check before treating a device as a therapy?
- Identify the product. A microneedle patch or nanostructure used in research is not automatically a medicine authorized for treatment.
- Check the payload and route. Delivery behavior depends on the formulation, device design, and intended tissue; evidence for one combination does not validate another.
- Look at the evidence stage. Laboratory or preclinical results do not show that patients will have the same outcomes. Trial activity is also not the same as approval.
- Check the relevant regulator and location. Regulatory status is product-specific; a nanomaterial’s presence by itself does not establish safety, effectiveness, or authorization.
