Earthworms can process leaves loaded with tannins and other polyphenols because their guts contain drilodefensins, a family of surface-active metabolites. In laboratory tests, these compounds reduced the protein precipitation caused by plant polyphenols without suppressing a tested earthworm digestive enzyme. That protection helps explain how earthworms handle chemically defended litter, but it does not make every leaf edible or by itself explain all decomposition in soil.
Why tannin-rich leaves are difficult to digest
Fresh leaves and fallen litter contain polyphenols, including tannins. These compounds help plants deter herbivores and microbes, but they also create a biochemical problem for any animal trying to digest the tissue. Polyphenols can bind soluble proteins and precipitate them into insoluble complexes. Digestive enzymes are proteins, so this reaction can remove enzymes from solution or interfere with their activity.
The 2015 study by Liebeke and colleagues reports that plant polyphenol concentrations can range from 1% to 25% of leaf dry weight, a range drawn from earlier botanical research rather than measured across all leaves. The same paper cites estimates that throughfall and leaching transfer more than 200 kilograms of soluble polyphenols per hectare each year into soil. Those quantities help explain why litter chemistry matters to decomposers.
The scale of the potential process is large: the study’s introduction cites an estimate of 3.5 × 1010 tonnes of terrestrial leaf litter turned over worldwide each year. That is a cited global estimate, not a new measurement from the earthworm experiments. The key question is how an earthworm’s digestive system keeps enzymes working while it consumes this chemically challenging material.

Drilodefensins provide a chemical defense for the earthworm gut
Liebeke et al. identified a group of earthworm-associated dialkylfuransulfonate metabolites and named them drilodefensins. The major compound was identified as 2-hexyl-5-ethyl-furan-3-sulfonate. It was found in all 14 earthworm species from three families examined in the study. The compound was concentrated in the gut, particularly the foregut, where incoming plant material first encounters digestive secretions.
The researchers also proposed related compounds numbered 3 through 6 from mass-spectrometry data. Those structures were putative: the paper did not report associated nuclear magnetic resonance data for them. They should therefore be treated as proposed members of the metabolite family, not as structures characterized with the same level of confirmation as the major compound.
Drilodefensins showed surface-active behavior, meaning they can act somewhat like biological surfactants. A surfactant has both water-compatible and less-water-compatible parts, allowing it to interact with interfaces and with substances that would otherwise clump or separate. In the earthworm gut, that chemistry appears to reduce the tendency of polyphenols to pull proteins out of solution.

The authors summarized the finding this way:
“These compounds counteract the inhibitory effects of polyphenols on earthworm gut enzymes, and high-polyphenol diets increase drilodefensin concentrations in both laboratory and field populations.”
The statement describes the experiments’ conclusion. It does not mean that drilodefensins neutralize every plant defense, guarantee digestion of every species of leaf, or replace the contributions of microbes and other decomposers.

What happens during digestion
1. Polyphenol-rich litter enters the foregut
An earthworm ingests partly decomposed leaves and soil. The foregut contains the highest concentration of the major drilodefensin reported in the study, placing the metabolite where plant polyphenols first mix with gut fluids and enzymes.
2. Drilodefensins interact with the polyphenol-protein mixture
In the study’s in-vitro tests, the metabolites reduced the protein precipitation produced by polyphenols. The practical implication is that more protein can remain available in the digestive fluid instead of being locked into an insoluble aggregate.
3. Digestive enzyme activity is less vulnerable
The tested earthworm gut enzyme retained its activity in the presence of the metabolite. This is an important distinction: the compound reduced the polyphenol-driven precipitation reaction without acting as an inhibitor of that enzyme in the assay.
4. The chemistry supports, rather than completes, decomposition
Once enzymes can work, they still must break down plant polymers, and microbes continue processing material before and after it passes through an earthworm. The drilodefensin mechanism is therefore best understood as a biochemical adaptation that removes one obstacle to digestion, not as a complete explanation for litter disappearance.
Evidence that earthworms adjust the metabolite to their diet
The researchers tested whether diet changed the amount of the major drilodefensin. In a two-week feeding experiment, they gave earthworms diets containing tannic acid or oak leaves, both rich in polyphenols. Each treatment used 10 earthworms. The major metabolite increased by about 50%, with P < 0.01 in the reported comparison.

A high-fat diet did not produce a significant increase. That contrast matters because it suggests the response was associated with polyphenol exposure rather than a nonspecific reaction to any nutritionally unusual food. The result is a treatment-specific observation from a short laboratory experiment, not a universal percentage for all earthworm populations or feeding conditions.

| Oak-leaf diet, two weeks | About 50% increase in the major metabolite; P < 0.01 | Supports an association with high-polyphenol food |
| High-fat diet | No significant increase | Does not show the same response to a non-polyphenol dietary change |
| Field-collected woodland worms | Higher levels than worms from grassland and mine sites | Pattern observed across sampled habitats; it is not a controlled habitat experiment |
Field samples added a second line of evidence. Worms collected from woodland sites had higher levels of the major metabolite than worms from grassland and mine sites. Woodland litter often contains chemically defended leaves, but the field comparison cannot isolate diet from temperature, soil chemistry, species composition, age, or other habitat differences. It supports a relationship between environmental exposure and metabolite abundance while leaving the exact causes unresolved.
How the gut mechanism relates to litter decay
Drilodefensins explain a potential advantage at the point of digestion. Broader studies measure what happens to litter and soil, and those outcomes should not be treated as interchangeable with enzyme protection.
Global evidence for faster litter mass loss
A 2020 global meta-analysis by Huang, González and Zou combined 340 observations from 69 independent studies. On average, the presence of earthworms doubled litter mass loss. The size and direction of effects varied with earthworm functional group and diversity, so “earthworms” is not a single ecological treatment. Different species differ in burrowing behavior, feeding location, litter preference and interactions with microbes.
The meta-analysis also found that effects on soil organic carbon were not significant across all observations. Faster disappearance of material from the litter layer does not automatically mean that more carbon is stored in soil. Carbon can be respired as carbon dioxide, moved downward, incorporated into microbial biomass, or redistributed among soil pools.

A forest field comparison
In a 2006 study of two sites in a northern hardwood forest in southcentral New York, reference plots contained 1.7 to 3 times more litter after 540 days than plots invaded by exotic earthworms. The study also reported site-specific differences among earthworm communities. This result shows that earthworm presence can strongly alter litter disappearance at particular sites; it does not provide a universal decay rate for every forest or species assemblage.
An 80-day oak-litter microcosm
A 2026 Applied Soil Ecology experiment used labeled Quercus serrata litter and Amynthas earthworms in microcosms. Over 80 days, the treatment produced 2.5-fold cumulative soil carbon-dioxide efflux, 27% more litter-derived carbon and 48% more litter-derived nitrogen incorporated into the 0–10 centimeter soil layer.
Those measurements describe carbon and nitrogen pathways in one controlled setup. They do not establish that all earthworms increase soil carbon dioxide by 2.5-fold, that all forests receive 27% more litter-derived carbon, or that the same changes persist over years. The experiment is useful because it separates several fates of litter-derived elements instead of treating “decomposition” as one number.

What the drilodefensin study does not yet answer
The 2015 work established the protective behavior in biochemical assays, but several steps in the complete digestive pathway remain open. The researchers specifically noted uncertainty about what happens to drilodefensin–polyphenol complexes later in digestion. It is not yet clear whether the polyphenols are released for excretion, whether drilodefensins are recycled, or whether enzymes degrade the metabolites.
Do these 3 things before closing this tab:
1Repair Windows errors before they cause bigger problems2Fix the driver behind crashes, sound loss and screen glitches3Clear out junk files and repair common Windows errorsThe experiments also do not show that drilodefensins are the only adaptation involved. Earthworm species differ in gut anatomy, microbial partners, feeding habits and tolerance of particular leaf chemistries. A leaf’s fiber content, nitrogen level, lignin concentration, moisture and stage of decay can change digestibility independently of polyphenols.

Finally, a higher metabolite concentration is evidence of a response, not proof of a complete cause-and-effect chain from diet to ecosystem carbon storage. Connecting gut chemistry to ecosystem outcomes requires measurements across species, litter types, soils, climates and timescales.
How to read the headline accurately
“Unique compounds let earthworms finish off leaves” is a shorthand for a narrower finding: earthworm guts contain drilodefensins that can counteract polyphenol-driven protein precipitation in laboratory tests, and earthworms exposed to polyphenol-rich diets produced more of the major metabolite in the reported experiments.
- Established: plant polyphenols can precipitate proteins; drilodefensins are gut-localized, surface-active metabolites; and the major compound reduced precipitation without reducing the tested enzyme’s activity in vitro.
- Supported by the feeding experiment: tannic acid and oak leaves increased the major metabolite by about 50% over two weeks, while a high-fat diet did not significantly increase it.
- Supported by field sampling: woodland worms had higher levels than grassland and mine-site worms.
- Still conditional: the magnitude of litter decay, carbon release and nutrient incorporation depends on species, functional group, litter quality, habitat, duration and the measured endpoint.
The biochemical adaptation gives earthworms a way to keep digestive proteins usable in the presence of plant chemical defenses. Microbes, physical fragmentation, soil movement and the subsequent fate of carbon and nitrogen determine what happens beyond that gut-level advantage.
