A 2017 spectroscopy study found that water next to DNA can produce a chiral vibrational signal: its measured O–H response bears the handedness of the DNA helix. The authors called the proposed arrangement a “chiral spine of hydration.” In practical terms, the result means that at least some water molecules in DNA’s hydration shell are organized in a way that follows the molecule’s twist.
That wording needs an important qualification. The researchers did not photograph a continuous column of water inside living cells. They measured a surface-sensitive optical response from water at a DNA interface, then interpreted the response with molecular models. The biological function of the arrangement remains unknown.
What “chiral spine of hydration” means
Chiral describes an object or arrangement that has a handedness: a right-handed version and a left-handed mirror image cannot be superimposed. The familiar DNA double helix is chiral. A DNA molecule therefore creates a three-dimensional environment that twists in one direction along its grooves rather than presenting a flat, mirror-symmetric surface.

Hydrogen bonds continually form and break, and the nearby bases, sugar-phosphate backbone, grooves and ions influence which orientations are more favorable. The study asked whether that local hydration environment could carry DNA’s handedness strongly enough to produce a chiral optical signal.

The phrase “spine” is a structural metaphor for a recurring, helix-following organization in the hydration shell. It should not be read as proof of a rigid rod or an unbroken water filament. The experiment detected a collective spectroscopic response from interfacial water; it did not resolve every molecule or establish that one continuous structure persists through a cell.
How the researchers tested the idea
Megan L. McDermott, Hannah Vanselous, Steven A. Corcelli and Poul B. Petersen used chiral sum-frequency generation (SFG) vibrational spectroscopy. SFG combines two laser fields at a surface or interface. When their frequencies add to a molecular vibration, the emitted signal reports on the vibrational modes of molecules that are arranged asymmetrically at that boundary.
In this work, the relevant vibration was the stretch of the oxygen–hydrogen (O–H) bond in water. The researchers recorded chiral SFG spectra, which are sensitive to handed arrangements, and compared them with achiral SFG spectra that report more broadly on the hydration response. The comparison is important: a chiral spectrum isolates the part of the interfacial response that follows a handed arrangement, while the achiral spectrum includes the larger, non-chiral background from water around DNA.
SFG is especially useful here because it is interface-selective and does not require fluorescent or chemical labels on the DNA or water. It can therefore probe a hydration layer without attaching a marker that might alter the structure being studied. It is still an indirect measurement: the spectrum must be interpreted using vibrational assignments and molecular simulations or models rather than read as a literal image.

The samples and conditions
| Experimental feature | What the study used | Why it matters |
|---|---|---|
| DNA design | Self-complementary, surface-bound 24-base-pair duplexes | A defined, reproducible DNA interface could be examined optically. |
| Sequences | (GC)12 and (AT)12, meaning 12 alternating GC or AT base pairs | Testing both designs shows the signal was not limited to one of the two sequences. |
| Solution | 100 mM sodium chloride in water | The paper describes this salt level as near-physiological conditions, while it remains a simplified laboratory solution. |
| Temperature | Room temperature | The result applies to the reported laboratory conditions, not automatically to every cellular environment. |
| Readout | Chiral and achiral SFG spectra of the water O–H stretch | The spectral contrast was used to infer differences in hydrogen bonding and molecular organization. |
The paper was received on March 6, 2017, published online on May 24, 2017, and appeared in the July 26, 2017 issue of ACS Central Science. Those dates identify the original report; they do not imply that the biological interpretation has since been settled.
What the spectra showed
A chiral response for both DNA sequences
Both (GC)12 and (AT)12 produced a chiral water response. The authors interpreted that shared result as evidence that DNA-templated chiral hydration is not confined to one of the two tested sequence designs. It is a narrower conclusion than saying every DNA sequence has the same feature. Only these two designed, surface-bound duplexes were examined in the reported experiment.
The chiral signal was shifted to lower vibrational frequencies
Relative to the broader achiral hydration response, the chiral O–H signal was shifted toward lower vibrational frequencies. In vibrational spectroscopy, a lower O–H stretching frequency is commonly associated with stronger hydrogen bonding because the hydrogen bond weakens the covalent O–H bond slightly and changes its vibrational energy.
The Tool Desk
Outbyte PC Repair FREERepair Windows errors before they cause bigger problemsFix Now →Outbyte Driver Updater FREEFix the driver behind crashes, sound loss and screen glitchesFind Drivers →Accordingly, the authors interpreted the shift as evidence that the water molecules contributing most strongly to the chiral response were, on average, more strongly hydrogen-bonded than the general population of water molecules detected in the achiral spectrum. This is an inference about an ensemble response, not a direct measurement of the force on each individual water molecule.

An additional feature in the AT sample
The (AT)12 spectrum also contained a feature the researchers attributed to non-hydrogen-bonded O–H groups. They suggested that tighter confinement in the AT minor groove could help explain why some water molecules appear in an environment where an O–H bond is not participating in a conventional hydrogen bond.
That explanation is plausible within the authors’ interpretation, but the spectrum does not show the individual water orientations as a photograph would. The assignment depends on how the vibrational feature is modeled and related to the geometry of the groove.
Where the “spine” is likely located
DNA has a major groove and a minor groove, and each groove presents a different arrangement of bases and chemical groups to surrounding water. The authors said the detected chiral response was likely dominated by water near the minor groove. The experiment could not completely exclude a contribution from water near the major groove, however.
This distinction matters because a statement such as “water in DNA forms a spine” can sound more precise than the measurement allows. The data support a chiral hydration response associated with the DNA interface, with the minor groove considered the most likely dominant source. They do not establish a single geometrically uniform track running through one groove, nor do they quantify the fraction of all nearby water that follows one orientation.

What the experiment establishes—and what it does not
Supported by the reported measurements
- Water around the two tested DNA duplexes generated a measurable chiral SFG response in the O–H stretching region.
- The response occurred for both the alternating GC and alternating AT 24-base-pair samples.
- The chiral response was displaced toward lower frequency compared with the broad achiral hydration response, consistent with stronger hydrogen bonding in the chiral population.
- The spectra can be used to interrogate DNA’s hydration environment without adding labels to the molecules.
- The authors’ molecular interpretation is consistent with DNA’s chirality being imprinted on nearby water.
Not established by this study
- It did not directly image a continuous water column or “spine.”
- It did not observe DNA inside living cells, nuclei or tissues.
- It did not test every DNA sequence, DNA form, salt composition, temperature or crowding condition.
- It did not demonstrate that the arrangement has a biological function.
- It did not show that the chiral hydration response controls gene expression, replication, folding or drug binding.
- It did not prove that the same structure persists unchanged when DNA is bound by proteins, ligands, ions or other biomolecules.
The authors explicitly stated that the biological relevance of a chiral spine of hydration is unknown. That caveat is part of the result, not a minor footnote. A spectroscopic signature can reveal an organized hydration environment without showing that the organization performs a useful task in an organism.

Hydration affects how DNA bends, fluctuates and presents its grooves to other molecules. Water and dissolved ions screen charges on the phosphate backbone, occupy pockets in the grooves and participate in the energetic trade-offs that accompany binding. A method that distinguishes a chiral component of this environment gives researchers another way to ask how DNA’s three-dimensional shape is communicated to surrounding solvent.
The value of the study is therefore methodological as well as structural. Chiral SFG can probe an interface without labels, and its sensitivity to O–H vibrations makes it possible to compare water populations that are otherwise difficult to isolate. The result opens a way to investigate whether hydration changes when DNA encounters different salts, proteins, small molecules or drugs.

How to interpret headlines about the discovery
A careful summary would say: Researchers detected a DNA-associated chiral water response with label-free SFG spectroscopy under controlled laboratory conditions. A headline that says “DNA has a water backbone” or “scientists photographed a water spine in cells” goes beyond the evidence.
Three details should travel with any explanation:
- Measurement type: the result is a vibrational spectrum from an interface, not a direct molecular image.
- Sample scope: the test used two surface-bound 24-base-pair duplexes, (GC)12 and (AT)12, in room-temperature 100 mM NaCl solution.
- Biological status: the function and relevance of the chiral hydration arrangement remain unknown.
Questions the study leaves open
Is the response universal across DNA?
The shared signal in the GC and AT samples suggests that the phenomenon is not restricted to those two sequence compositions. It does not establish universality. Longer or shorter DNA, mixed sequences, different helical forms, mismatches and DNA packaged with proteins could alter the hydration response.
Does the signal come only from the minor groove?
The minor groove was considered the likely dominant contributor, but major-groove water could not be ruled out completely. Separating the two contributions would require experiments or modeling with greater spatial or structural discrimination.

Pick the symptom - the matching free tool is one click away.
Does “stronger hydrogen bonding” mean the water is immobile?
No. A lower-frequency O–H response indicates a different hydrogen-bonding environment in the measured ensemble. Water molecules can still exchange partners and reorient rapidly. The spectrum alone does not provide a residence time or prove that the arrangement is rigid.
Could cells contain the same arrangement?
Cells contain DNA together with proteins, metabolites, ions, crowding agents and constantly changing mechanical conditions. The reported experiment did not reproduce that environment. It provides a testable physical possibility, not an in-cell observation.
Bottom line
The “chiral spine of hydration” is the authors’ name for a DNA-shaped organization inferred from a chiral water-vibration signal. Under the study’s controlled conditions, both tested DNA sequences produced that response, and its lower-frequency position was interpreted as stronger hydrogen bonding than in the broader hydration signal. The finding shows that DNA’s handed structure can be detected in the behavior of nearby water; it does not yet show a continuous water spine in living cells or a demonstrated biological function.
