A 2015 computational study found a strong correlation between predicted disorder in two Ebola proteins, VP30 and NP, and reported case-fatality rates across Ebola virus types. The authors proposed that disorder could help the virus replicate and interact with other molecules, but their analysis did not show that disorder causes higher lethality.
What “intrinsic disorder” means in a viral protein
Many proteins adopt a stable, well-defined three-dimensional structure. An intrinsically disordered protein, or a disordered region within one, does not maintain one fixed structure across conditions. That flexibility does not make it biologically inert: disordered regions can interact with other molecules and participate in cellular processes.
The 2015 study focused on the predicted percentage of disorder in two proteins associated with Ebola’s nucleocapsid, the structure that packages the virus’s genetic material: VP30 and nucleoprotein, or NP.
What the 2015 comparison found
Gerard Kian-Meng Goh, A. Keith Dunker, and Vladimir N. Uversky compared the percentage of disorder in VP30 and NP with reported case-fatality rates for Ebola virus types. They reported a high correlation between the disorder measurements and those rates. The paper appeared in Molecular BioSystems in 2015; PubMed’s record and abstract describe the comparison.

The abstract does not provide a numerical correlation coefficient or sample size, so those figures cannot be stated from that record. A correlation also cannot establish direction or cause: it does not show that greater disorder made a virus more lethal, nor rule out other differences among virus types.
Why the authors thought disorder might matter
The authors proposed that greater disorder could assist viral RNA transcription and allow broader protein binding. In their proposed account, those interactions might support viral multiplication and nucleocapsid functions. This is a possible explanation for the association, not a demonstrated sequence of events linking disorder to case fatality.

In a June 2015 report, co-author Vladimir Uversky said the correlation could open “new strategies for Ebola virus vaccine development.” That was a suggestion about a possible research direction—not evidence that the correlation produced a tested vaccine design or an approved vaccine. USF Health’s report attributes the statement to him.

A separate experiment offers molecular context
A different study examined a disordered peptide from Ebola’s VP35 protein, rather than comparing disorder across virus types. The peptide, NPBP (residues 20–48), bound NP; researchers solved the complex structure at 3.7 Å resolution and reported that the peptide inhibited NP oligomerization and released RNA from NP–RNA complexes in vitro. These results show a specific molecular interaction under experimental conditions, but they do not verify the strain-level correlation with case-fatality rates.

The VP35 work was published in Cell Reports on April 21, 2015. The open-access article provides the experimental details.
Do not confuse lethality with the scale of the 2014 outbreak
A separate computational analysis of Ebola sequence changes from 1976 to 2014 reported no evidence that amino-acid replacements explained the exceptional scale of the 2014 outbreak. It found many changes in poorly constrained, intrinsically disordered regions. That analysis addressed sequence changes and outbreak scale; it was not the 2015 comparison of disorder percentages in VP30 and NP with case-fatality rates. What the finding can—and cannot—tell us
- It can: identify an association that may help researchers form hypotheses about viral protein function and differences among Ebola virus types.
- It cannot: establish that intrinsic disorder causes greater lethality, quantify the relationship from the accessible abstract, or demonstrate a clinical intervention.
- It should be read alongside: molecular experiments such as the VP35 peptide study, while keeping those experiments distinct from cross-type statistical comparisons.
The title appeared in Chemistry World’s June 12, 2015 archive under the wording “Intrinsically disordered proteins may be vital in determining the deadliness of Ebola strains.” Its careful qualifier—“may”—fits the evidence: an intriguing computational association with proposed mechanisms, not a settled explanation of Ebola lethality.
