Researchers at Cornell University College of Veterinary Medicine and the Baker Institute for Animal Health have identified the first molecularly characterized case of Marfan syndrome in domestic cats. The affected cats, brothers named Gary and Shaggy, developed unusually long limbs as kittens. Later examinations found abnormalities in their eyes and enlargement of the aorta, leading veterinarians to investigate an inherited connective-tissue disorder.
What disease did Cornell find in the cats?
The cats had a feline form of Marfan syndrome, a disorder in which weakened connective tissue affects structures such as blood vessels, bones, ligaments, skin and the eyes. Cornell describes Gary and Shaggy as the first domestic cats with molecularly characterized Marfan syndrome.
Marfan syndrome is best known as a human condition, affecting about 1 in 4,000 people, according to Cornell University College of Veterinary Medicine in 2026. The feline cases are not evidence that cats commonly develop the disease; they are two related animals whose clinical signs and genetic results matched a Marfan-like disorder.
How veterinarians recognized the condition
Unusually long limbs
Gary and Shaggy stood out because of their long legs during kittenhood. Disproportionately long limbs can indicate abnormal connective-tissue development, but that sign alone cannot establish Marfan syndrome.

Eye abnormalities
Follow-up examinations identified structural abnormalities in the cats’ eyes. The eye findings strengthened the suspicion because fibrillin-1, the protein affected in Marfan syndrome, contributes to the support structures of the eye.
Aortic enlargement
Veterinarians also found enlargement of the aorta, the body’s main artery. A weakened aortic wall is one of the most serious features associated with Marfan syndrome because it can progress to life-threatening vascular complications.
The combination of limb differences, ocular abnormalities and aortic enlargement prompted genetic investigation rather than a diagnosis based only on appearance.

What is the FBN1 mutation in Gary and Shaggy?
Sequencing found changes in FBN1, the gene that encodes fibrillin-1. Fibrillin-1 is a structural component of connective tissue throughout the body, including blood vessels, bones, ligaments, skin and eyes.
Both brothers carried two altered copies of FBN1, one inherited from each parent. The paper describes the change as a homozygous hypomorphic splice-region variant. In practical terms, the variant interferes with the processing of the gene’s instructions but does not completely switch the gene off.
That partial activity matters. The cats retained enough normal fibrillin-1 function to survive into adulthood, unlike what might be expected from a variant that entirely eliminated the gene’s function.

How the feline finding differs from human Marfan syndrome

The inheritance difference does not mean the cats have a completely unrelated condition. It shows that the same connective-tissue gene can be disrupted through a different genetic pattern and molecular mechanism.

The peer-reviewed study, “A homozygous hypomorphic FBN1 splice region variant in domestic cats with Marfan syndrome,” was published in Scientific Reports in 2026 (DOI: 10.1038/s41598-026-70702-3). Cornell scientists worked with researchers at Ghent University in Belgium, the University of Pennsylvania and the Schwarzman Animal Medical Center in New York City.
Senior author Dr. Jacquelyn Evans, an assistant professor in Cornell’s Department of Biomedical and Translational Sciences and at the Baker Institute for Animal Health, said the findings “provide a foundation for improved veterinary diagnostics.” She added that the discovery may help veterinarians recognize similar cases and may support development of genetic tests.

The case also illustrates the value of comparative medicine. Studying naturally occurring disease in animals can reveal how a gene behaves in another species while improving care for the animals themselves. Evans said the work began with collaboration among pet parents, veterinary clinicians and genetic experts.

These cases show that domestic cats can develop a Marfan syndrome phenotype associated with FBN1 variants. They do not establish how common it is, whether the same variant occurs in other cat populations or whether every cat with long limbs has the syndrome.

A cat with unusual proportions would need a veterinary examination and appropriate imaging before anyone could assess possible connective-tissue disease. Long limbs by themselves are not diagnostic, and genetic results must be interpreted alongside physical findings and family history.

Could this lead to a genetic test for cats?
Possibly, but no commercial feline test or validated treatment was established by this report. The identified variant gives researchers a defined target for future testing and may help veterinarians recognize related cases. Any test would need validation in additional cats to determine its accuracy, disease risk and relevance across breeds and families.
For now, the practical advance is diagnostic awareness: veterinarians who see the combination of disproportionate limbs, eye changes and aortic enlargement can consider an inherited connective-tissue disorder and pursue specialist evaluation.
