Click chemistry can join modified DNA oligonucleotides through a triazole-containing backbone link, and one 2014 study showed that human cells could transcribe a reporter gene containing a single such link. That is a specific proof of concept—not evidence that click chemistry makes complete gene synthesis faster overall or that genes with many click links will work.

What a click-linked DNA strand is

In this approach, short DNA oligonucleotides are prepared with complementary reactive handles: an azide on one strand and an alkyne on another. Copper-catalyzed azide–alkyne cycloaddition (CuAAC) joins them, forming a triazole-containing linker at the junction. The backbone at that point is therefore non-natural rather than the usual phosphodiester connection.

This is a backbone modification, not an expanded genetic alphabet: the unusual feature is the connection between nucleotides, not a new base-pairing scheme. In template-directed ligation, a DNA template or splint positions the modified oligonucleotides in the intended order before reaction.

What experiments have demonstrated

Joining and copying modified DNA

Work published in 2007 demonstrated CuAAC-mediated, template-directed joining of oligonucleotides bearing 5′-alkyne and 3′-azide groups. It also described template-free oligonucleotide circularization and catenation. A later study showed PCR amplification through a triazole linkage, establishing that a polymerase could copy across that junction in the tested setting. These results established useful precursor capabilities, not a complete, broadly validated gene-synthesis workflow. 2007 template-directed ligation study; 2009 PCR amplification study.

Transcription of a click-linked reporter gene

In 2014, researchers tested a click-linked gene encoding mCherry in human cells. They sequenced cDNA derived from the resulting mRNA and reported error-free transcription for that construct. This supports the conclusion that cellular transcription machinery tolerated the particular triazole junction in the experiment; it does not establish that every sequence or arrangement of unnatural links will be transcribed correctly. Read the 2014 study.

The tested scope is narrow

The demonstrated cellular result involved a gene with a single click linkage. It does not show that genes carrying many such junctions function normally, nor does it settle how performance changes with the number or placement of links. Chemistry World’s 2014 coverage quoted Geoff Baldwin noting that the effect of ten linkages in one gene was unknown at the time; that was a contemporaneous caution, not a later resolution of the question. Does click chemistry make gene synthesis faster?

Not on the evidence available here. CuAAC offers a chemical way to connect suitably modified oligonucleotides, and the reaction has been discussed as a route for assembling DNA. But the cited work does not report a head-to-head comparison of total end-to-end synthesis time against enzymatic gene assembly or other assembly methods. Reaction efficiency is not the same as proof of a faster overall workflow: oligo preparation, alignment, reaction, purification, verification and assembly all contribute to the total process.

The 2014 paper framed the motivation this way: “The practical limit on the length of error-free oligonucleotide synthesis has necessitated the use of enzymes for the assembly of polynucleotide chains into genes.” That motivates an alternative chemical assembly route; it does not demonstrate a general speed advantage. The article’s headline framing should therefore be read as a proposal or possibility, not a measured comparison.

What a methods comparison should examine

There is no unified head-to-head performance dataset in these sources from which to rank click ligation, enzymatic assembly and other chemical ligation routes numerically. A fair evaluation would compare the following:

  • Biological read-through: whether polymerases can copy across the junction and whether the resulting transcript or gene functions in the tested system.
  • Linkage scope: how many non-natural junctions were tested, where they occur, and whether the evidence is biochemical, cellular or functional.
  • Oligo design: which azide and alkyne end groups are required, whether a template or splint is needed, and how the oligonucleotides are ordered.
  • Reaction conditions: whether the method uses copper-catalyzed or copper-free chemistry and what conditions the specific protocol requires.
  • Workflow burden: reaction yield, purification, read-through and verification requirements. A 2022 study in a sequencing-related context highlights purification and low read-through efficiency as possible workflow constraints; that application is not the same as the 2014 gene experiment, so it does not directly negate its result. Read the 2022 study.

A 2012 review discusses splint-based organization, chemistry requirements and limitations relevant to robust chemical DNA assembly. Those design and process details matter when assessing whether a ligation method is useful for a particular construct. Read the review.

What this means for practical use

Click ligation is a specialized research method for joining oligonucleotides that carry the required reactive groups. Specialist suppliers list related modified supports and custom oligonucleotide services, but a catalog listing alone does not establish that the exact end groups and chemistry used in the 2014 experiment are currently available as a ready-to-order product. Researchers evaluating a supplier should verify the precise modification, sequence, scale, purity and quality-control options directly.

The result is scientifically meaningful but bounded: one click-linked mCherry gene was transcribed accurately in human cells in the reported experiment. Whether multiple triazole links can be used reliably, and whether the complete workflow can outperform conventional assembly in time or throughput, are separate questions not answered by that result.
