To speed up chiral catalyst screening without losing useful information, parallelize the experiments, choose an analytical method that measures the outcome you care about, and use data or models to decide what to test next—not to replace experimental confirmation. High-throughput reaction execution only helps if the readout can also distinguish activity from enantioselectivity and the results hold across the substrates you intend to use.
What makes chiral catalyst screening slow—and easy to misread?
A screen explores more than a list of catalysts. The catalyst or ligand, substrate, solvent, temperature, pressure, additives and other conditions can interact. A small change may affect reaction activity, enantioselectivity, or both. Optimizing one variable at a time can therefore miss combinations that perform well together.
There is also an analytical bottleneck: measuring conversion or product formation is not the same as measuring enantiomeric excess (ee), the measure of how strongly a reaction favors one enantiomer over the other. If selectivity is the goal, a fast activity screen alone cannot identify the best catalyst. The 2026 review by Yu and Zhao surveys chiral-analysis approaches including chromatography, spectroscopy, mass spectrometry and NMR, underscoring that the right readout depends on the reaction and sample (review record).

Which faster screening methods fit which parts of the job?
These approaches can complement one another, but their reported figures describe specific studies rather than universal benchmarks. In particular, a rate for analysis is not necessarily the rate for running, preparing and analyzing an entire screen.

| Approach | What it speeds up | Demonstrated evidence | What to check before applying it |
|---|---|---|---|
| Parallel high-throughput experiments (HTE) | Reaction execution across catalyst and condition combinations | Kalikadien et al. report screening 192 Rh precatalysts against five model substrates and routinely testing two 96-member ligand plates with Rh. Their Rh-catalyzed asymmetric olefin hydrogenation dataset contained up to 3,552 data points (Chemical Science study, 2024). | The study is specific to Rh-catalyzed asymmetric hydrogenation. Its solvent results also show why conditions matter: solvent had a particularly significant effect on performance, mainly activity and to a lesser extent ee. |
| Ion-mobility mass spectrometry (IM-MS) with diastereoisomerization | Ee analysis and mapping a large reaction set | Nie et al. report approximately 1,000 reactions analyzed per day, a median ee-analysis error below ±1%, and mapping more than 1,600 reactions in their demonstration (Nature Communications study, 2023). | These are figures for the demonstrated method and study, not a promise of end-to-end screening throughput or accuracy on every substrate, instrument or crude-mixture matrix. Check analytical compatibility and validation for your own samples. |
| Multi-substrate HTE with NMR and machine learning (ML) | Optimizing conditions while accounting for more than one substrate | Seo et al. report an eightfold faster workflow for their asymmetric mono-reduction of 1,2-dicarbonyls, using simultaneous quantitative NMR analyses and ML (Angewandte Chemie International Edition article record, first published 2025). | The speedup belongs to that workflow and reaction class; it is not a general benchmark for catalyst screening. Confirm that the NMR readout and optimization target suit your reaction. |
| Virtual catalyst screening | Narrowing which catalyst candidates to test experimentally | CatVS reports virtual screening for asymmetric transformations, with selected ligand/substrate combinations validated experimentally (Nature Catalysis study, 2018). | Predictions and candidate rankings are system-dependent. Experimental validation is necessary, and results should not be transferred automatically to unrelated catalyst families. |
| ML trained on reaction data | Prioritizing experiments within a represented chemical domain | The Rh hydrogenation study evaluates prediction from high-throughput data and identifies continuing challenges in predicting enantioselectivity (Chemical Science study, 2024). | Performance can fall outside the model’s training domain. The study cautions against over-interpreting small or narrow datasets; a prediction is a way to prioritize a test, not experimental proof. |
How should you design a faster screen?
-
Choose the decision the screen must support
Write down whether the priority is activity or conversion, ee, regioselectivity, robustness across substrates, or a balance of outcomes. Define the assay limits and how you will rank candidates before collecting results. Otherwise, a screen can produce a large dataset without resolving which catalyst is actually preferable.
-
Build a chemically meaningful experiment set
Select catalyst or ligand diversity and reaction conditions relevant to the intended chemistry. Parallel formats can expand the combinations tested; the Rh hydrogenation work demonstrates two 96-member ligand plates, but that does not establish that every reaction is compatible with the same setup. If using 96-well microplates, match plate material, sealing, well geometry, temperature tolerance and solvent compatibility to the chemistry.

-
Match the readout to the objective
Measure reaction outcome and selectivity when both matter. An activity or conversion signal cannot stand in for ee. Choose an analytical method that can handle the substrate, crude-mixture matrix and required sensitivity at the intended throughput. IM-MS is one demonstrated route to rapid ee analysis, not a universal replacement for chromatography or other methods.

Record conditions and failures consistently
Keep catalyst identity, substrate, solvent, temperature, additives and other tested conditions attached to each result, including failed or low-performing reactions. Consistent records help distinguish a real catalyst effect from a condition change and provide more useful data if you later build or apply a predictive model.

-
Use predictions to select the next experiments
Apply ML or virtual-screening rankings when the candidates and conditions are sufficiently represented by the data or calculations behind them. Treat uncertain or out-of-domain predictions as hypotheses to test. The 2022 RIKEN summary describes one computational-screening example in asymmetric N-heterocyclic carbene-copper catalysis using 30 training samples; that specific example does not establish a generally sufficient training-set size (RIKEN research summary).
-
Check generality before optimizing only one model substrate
If the intended reaction must work on a substrate class, include more than one relevant substrate early enough to reveal trade-offs. A catalyst that ranks highly on a single model compound may not retain its activity or selectivity elsewhere. Seo et al.’s multi-substrate workflow is a specific example of using this broader objective, not proof that the same acceleration will transfer to other transformations.

-
Re-test promising conditions
Repeat selected results and assess reproducibility before treating a lead as robust. A model-ranked candidate or a single high-throughput measurement is a reason to confirm, not a substitute for confirmation.

How can you tell whether a speed claim applies to your screen?
Before adopting a published throughput figure, identify which stage it measures. A reported number of reactions analyzed per day may describe the analytical method, not reaction setup, execution, sample preparation and analysis together. Likewise, a reported workflow acceleration applies to its demonstrated reaction and operating setup unless broader validation is shown.
