Short answer: The phrase “DNA to direct and switch off chemo” describes a 2009 laboratory drug-delivery experiment, not a treatment patients can use. Zehui Cao and colleagues attached a nucleolin-targeting DNA aptamer to liposomes containing cisplatin. In the cell-delivery system, adding complementary DNA was reported to control how much of the payload was delivered.
That “switch off” was a control over an experimental delivery mechanism. It did not neutralize cisplatin already acting in a body, reverse chemotherapy side effects, or give patients a way to stop prescribed treatment. The available reports do not establish human safety, clinical benefit, regulatory approval or current availability.
What the 2009 headline actually referred to
The exact headline appeared in a Chemistry World news report dated 28 July 2009. Its wording made the idea sound like a remote control for chemotherapy: DNA would guide a drug toward cancer cells and then stop delivery if toxicity appeared. The underlying work was narrower. It described a cell-targeted delivery vehicle tested experimentally, not a clinical protocol.
The closest matching primary study, by Zehui Cao and colleagues, used liposomes as microscopic carriers. The liposomes contained cisplatin, a cytotoxic chemotherapy drug. A DNA aptamer was attached to the carrier so the system could recognize a chosen cellular target, identified in the study as nucleolin. Complementary DNA was then used as an “antidote” to regulate the extent of delivery in the experimental setup.

Those details matter because “switch off chemo” can mean several different things. In this case it meant reducing or controlling further delivery from a designed carrier. It did not mean removing cisplatin from the bloodstream, undoing damage to healthy tissue, or replacing a doctor’s decision about whether treatment should continue.
How the aptamer–liposome system was designed to work
- Load the carrier. Cisplatin was placed inside a liposome. A liposome is a membrane-bound particle used experimentally to transport a drug; it is not the same thing as free cisplatin circulating on its own.
- Add the targeting strand. The liposome was conjugated to a DNA aptamer. Aptamers are short nucleic-acid strands selected for their ability to bind a particular molecular target. Here, the aptamer provided the cell-targeting function, while the liposome provided the container.
- Expose cells to the construct. The reported work examined delivery to cells in an experimental setting. The abstract describes control of delivery extent, but it does not present a patient treatment schedule or clinical outcome.
- Introduce complementary DNA. A strand designed to pair with the aptamer was added. The study described this complementary strand as an antidote because it could alter the aptamer’s active configuration and thereby control how much carrier-mediated delivery occurred.
The division of labor is easy to miss in a short headline: the aptamer was the recognition element, the liposome was the carrier, cisplatin was the payload and the complementary strand was the control signal. DNA did not replace the chemotherapy drug or make cisplatin harmless.
What “switch off” means—and what it does not
What the experiment suggests
If a delivery system can be modulated after it reaches a target, investigators may be able to limit additional payload release or uptake. That is the practical meaning of the control described in the abstract. It is a property of the engineered carrier-and-DNA combination, demonstrated in an experimental cell-delivery context.
What it does not demonstrate
- It does not show that complementary DNA can deactivate cisplatin molecules that have already entered cells or begun damaging DNA.
- It does not show that an adverse reaction in a patient could be reversed on demand.
- It does not establish that every carrier particle would stop responding at the same time or that drug exposure could be reduced to zero.
- It does not provide instructions for changing, pausing or cancelling a prescribed chemotherapy regimen.
Anyone receiving chemotherapy should treat new or worsening symptoms as a medical issue and contact the oncology team or emergency services according to the instructions for that treatment. The 2009 delivery concept is not a substitute for those decisions.

What evidence was reported
The primary abstract describes cell-specific cisplatin delivery using a nucleolin aptamer-functionalized liposome and says that complementary DNA could control the extent of delivery. It does not, in the material available for this article, establish a human trial, a survival advantage, a validated toxicity reduction, or a dose that clinicians could prescribe.
The Chemistry World item is therefore best read as a report about an early-stage platform. Its “if unwanted side effects appear” wording explains the motivation for an adjustable carrier, but it should not be read as proof that side effects were safely switched off in patients. No clinical population, response rate, adverse-event rate or approved indication is supplied.
Translation from a cell experiment to medicine would require evidence about distribution through the body, stability of both DNA strands, immune reactions, how the carrier reaches a tumor, how much cisplatin reaches healthy organs, and how quickly the control strand acts. The cited reports do not resolve those questions.
How this platform differs from other 2009 delivery ideas
Several targeted-delivery approaches were being explored around the same time. They should not be merged into one technology or treated as competing clinical treatments. The following comparison is limited to the mechanisms and payloads described in the cited reports.

| Experimental platform | Payload | Targeting or release control | What the report establishes |
|---|---|---|---|
| Aptamer-functionalized liposome | Cisplatin | Nucleolin aptamer for cell targeting; complementary DNA reported to control delivery extent | Experimental cell delivery; no clinical protocol or patient benefit established |
| Targeted minicells | siRNA or a cytotoxic drug | Targeted minicells used for sequential treatment of drug-resistant tumors | A separate Nature Biotechnology study dated 28 June 2009; not the same carrier or DNA-control mechanism |
| pH-responsive carbon nanotubes | Doxorubicin | Folic-acid targeting with lower pH used to trigger release | A separate Biomaterials report describing delivery experiments in HeLa cells; not evidence for the aptamer-liposome system’s clinical status |
The table shows why a broad statement such as “DNA switched off chemotherapy” is misleading. Only the first row involves the complementary-DNA control described in the headline’s closest matching study. The minicell and carbon-nanotube papers used different materials, payloads and control mechanisms.
Why a controllable carrier could be useful in principle
Cisplatin is effective because it is toxic to dividing cells, but toxicity is also the central challenge of using it. A carrier that concentrates exposure near a selected cell type could, in principle, change the balance between tumor exposure and exposure elsewhere. A second molecular input that limits further delivery could, in principle, add an extra layer of timing control.
Those are design goals, not demonstrated patient outcomes. A targeting ligand can bind unintended cells, a carrier can break down before reaching its destination, and a control strand can have limited access to particles or tissues. Even if a carrier stops releasing new drug, molecules already delivered may continue to act. The 2009 report does not quantify these effects.

Why an “antidote” is not the same as a chemotherapy antidote
The word “antidote” in the abstract refers to the complementary DNA strand’s role in the engineered delivery circuit. It is a molecular counterpart to the aptamer, not a general antidote to cisplatin poisoning. The strand changes the behavior of the targeting or delivery component; it is not described as binding and neutralizing all cisplatin in a patient.

This distinction also explains why the headline’s promise can be overstated. A true clinical reversal would require a reliable way to halt exposure throughout the body and to treat injury that had already occurred. Neither outcome is shown by the cell-delivery description.
Historical timeline
- 28 June 2009: A Nature Biotechnology paper examined sequential treatment of drug-resistant tumors with targeted minicells carrying siRNA or a cytotoxic drug. This was a different delivery platform.
- 2009: A Biomaterials paper described doxorubicin-loaded single-wall carbon nanotubes, using folic acid for targeting and lower pH to trigger release in delivery experiments with HeLa cells. This was also distinct from the aptamer-liposome system.
- 28 July 2009: Chemistry World published the exact-title report “DNA to direct and switch off chemo,” summarizing the idea of controllable, targeted delivery.
The date is important: this was an early wave of nanomedicine and nucleic-acid delivery work. A 2009 news report should not be presented as a current announcement of an available therapy.

How to interpret similar headlines
When a story says that DNA can “direct” a drug, ask which part of the system DNA performs. It may be a targeting ligand, a structural component, a sensor or a switch. In this case it was the aptamer and its complementary control strand; the drug itself remained cisplatin and the container remained a liposome.
When a story says treatment can be “switched off,” ask whether the report measured carrier release, drug concentration, biological activity or patient symptoms. Those are different endpoints. The 2009 report supports the first type of claim—control of delivery extent in an experimental system—not the latter three.
Finally, check the date and platform. A result involving minicells, carbon nanotubes, siRNA or doxorubicin cannot be used as proof that a cisplatin liposome controlled by complementary DNA works in humans. Similar goals do not make the technologies interchangeable.

DNA was used as part of a proposed control circuit for targeted cisplatin delivery in laboratory experiments reported in 2009. A nucleolin aptamer helped direct a liposome, and complementary DNA was reported to regulate the amount delivered. That is a promising engineering concept, but it is not evidence that patients can stop chemotherapy, reverse side effects or obtain an approved DNA-controlled treatment.
