Yes, but only in a tightly engineered laboratory system. Researchers made an Escherichia coli strain maintain DNA containing an unnatural base pair, then showed that the added information could be transcribed and used by ribosomes to make a protein. The result was not a naturally evolved bacterium with a self-sustaining new alphabet. It was a semi-synthetic organism whose engineered transport system imported the unusual nucleotide building blocks from its growth medium.
The distinction matters. The 2014 work demonstrated propagation of DNA carrying the extra pair. A 2017 follow-up demonstrated information retrieval: transcription, decoding at the ribosome and site-specific insertion of natural or non-canonical amino acids into superfolder green fluorescent protein (sfGFP). Together, the studies show that genetic information can be expanded beyond DNA’s usual four-letter alphabet under controlled conditions.
What the “unnatural DNA transplant” actually was
DNA in ordinary cells uses four bases—adenine, thymine, cytosine and guanine—arranged as two complementary pairs. The experiments added a third pair, called dNaM–dTPT3, to an engineered E. coli system. These bases were not part of the bacterium’s natural genome and were not produced by an evolutionary change inside the cell.

An engineered transporter moved those molecules into the cell, where the replication machinery could use them alongside the natural nucleotides. The added pair was placed in DNA carried by the engineered organism, allowing the researchers to test whether the information could be copied and, later, read out to make a protein.

That is why the most accurate description is “semi-synthetic organism.” Some of its genetic chemistry was biological, while the extra letters and the means of supplying them depended on laboratory engineering.
Why adding a base pair is difficult
A genetic alphabet is useful only if several linked processes work together. A cell must be able to import or synthesize the nucleotide triphosphates, insert the bases into DNA, copy them during replication, preserve their pairing rules, transcribe DNA into RNA and have the ribosome interpret the resulting codons. Failure at any stage would make the extra information disappear or become unreadable.
Natural base pairs are recognized by the enzymes that copy DNA. An unnatural pair therefore has to fit into the geometry and chemistry of those reactions well enough to be copied, while remaining different enough to carry new information. The dNaM–dTPT3 pair was selected for this role in the reported system. The reported system did not replace every base in the chromosome. It tested a deliberately placed pair in an engineered genetic context.
The 2014 breakthrough: DNA with the extra pair could propagate
The first major milestone was reported in 2014 in the Nature study A semi-synthetic organism with an expanded genetic alphabet. Researchers created an engineered E. coli strain that maintained DNA containing an unnatural base pair. “Propagation” here means that the DNA carrying the pair could be copied and retained through cell growth in the experimental setup.

The result established an important but limited capability: an organism could keep genetic material containing more than the normal four DNA letters. The experiment was a proof of biological compatibility, not yet proof that the new letters could direct production of a new protein.
The imported-nucleotide requirement was central to the design. The bacterium did not manufacture the unnatural triphosphates from ordinary cellular metabolites. Its transporter brought them in from the surrounding growth medium. Without that supply chain, the extra bases could not be treated as an independent, self-sustaining alphabet.
The 2017 advance: the added information could be read
A 2017 Nature paper by Zhang and colleagues, A semi-synthetic organism that stores and retrieves increased genetic information, extended the earlier result. The researchers showed that information encoded with the dNaM–dTPT3 pair could be transcribed into unnatural codons and decoded at the ribosome.
They used this system to direct site-specific incorporation of amino acids into sfGFP, a fluorescent reporter protein. The demonstrated amino acids included both ordinary amino acids and non-canonical amino acids—building blocks that are not part of the standard set used by most natural proteins. This experiment connected the extra DNA letters to an observable protein product.

The distinction between the two milestones is best expressed as storage versus retrieval. In 2014, the engineered cell propagated DNA containing the pair. In 2017, the system retrieved that information through transcription and translation. The later result therefore showed not just that the extra letters could remain in DNA, but that they could participate in a genetic instruction.
The 2017 paper’s authors wrote: “The resulting semi-synthetic organism both encodes and retrieves increased information and should serve as a platform for the creation of new life forms and functions.” That is a description of a research platform and its prospective uses, not a claim that a new commercial organism already exists.
How the laboratory system worked
- Design a DNA construct. Researchers placed the unnatural pair in DNA used by the engineered E. coli system, rather than attempting to rewrite every position in the bacterium’s genome.
- Engineer nucleotide import. The cells received a transporter capable of bringing the required unnatural nucleotide triphosphates into the cell from the culture medium.
- Provide the chemical building blocks. The growth environment contained the dNaM and dTPT3 triphosphate forms needed for DNA synthesis. The cell’s natural machinery could then encounter the extra substrates during replication.
- Test propagation. In the 2014 stage, scientists checked whether DNA containing the pair could be maintained as the engineered cells grew. This established copying and retention in the reported experimental context.
- Test transcription. In the follow-up work, the sequence containing the added letters was transcribed so that the information entered the RNA-based stage of gene expression.
- Test ribosomal decoding. The resulting unnatural codons were presented to the translation system. The researchers measured whether the ribosome-linked machinery could place specified amino acids at the intended site.
- Observe a protein product. sfGFP served as a reporter. Successful site-specific incorporation produced a direct test of whether the extra genetic information had been converted into a protein sequence.
2014 and 2017 compared
| Milestone | Primary question | What was demonstrated | Role of supplied chemicals |
|---|---|---|---|
| 2014 Nature report | Can engineered bacteria maintain DNA with an added base pair? | Propagation of DNA containing an unnatural pair in an engineered E. coli strain. | Unnatural nucleotide triphosphates had to be imported from the growth medium. |
| 2017 Nature follow-up | Can the added sequence carry a usable genetic instruction? | Transcription into unnatural codons, ribosomal decoding and site-specific amino-acid incorporation in sfGFP. | The same semi-synthetic design supplied the chemical inputs needed for the added alphabet. |
This comparison prevents a common overstatement. Saying that the 2014 bacterium “made a protein from unnatural DNA” collapses two separate achievements. Protein decoding was the later milestone.

What the experiment proves—and what it does not
What it proves
- An engineered bacterial system can maintain DNA containing a third base pair in addition to the four natural bases.
- The dNaM–dTPT3 pair can carry information that reaches the transcription and translation machinery in the reported setup.
- A genetic instruction using the added information can direct site-specific incorporation of selected amino acids into a reporter protein.
- Genetic alphabets are not restricted to four letters by an absolute chemical impossibility; additional symbols can work when the supporting biology is engineered around them.
What it does not prove
- It does not show that an ordinary wild E. coli strain can spontaneously acquire and use the extra pair.
- It does not show that the bacterium evolved a new alphabet independently.
- It does not show that the organism synthesizes all of the unnatural nucleotide triphosphates by itself.
- It does not amount to replacing the entire natural genome with an alternative alphabet.
- It does not establish a consumer product, medical treatment or industrial process ready for routine use.
Why an expanded alphabet matters
Four DNA letters provide three-letter codons that are already used extensively by natural biology. Adding another pair creates additional sequence symbols and can increase the information that a designed DNA segment carries. The 2017 protein experiment showed one practical consequence: the extra symbols can help specify amino acids that ordinary genetic coding does not normally assign at a chosen position.

That capability could support research into proteins with new chemical properties, biological sensors, molecular tags or other functions that depend on amino acids outside the standard set. It could also give synthetic biologists a larger design space for separating engineered instructions from the natural genetic code.
Those are prospective applications. The papers establish a platform for exploring such functions, not a finished technology with demonstrated commercial performance. Any real-world application would still have to address stability, manufacturing, containment, reproducibility and the need to control the supply of the unnatural building blocks.

Dependence on laboratory support
The organism’s extra alphabet was conditional on the engineered transport pathway and the medium containing the required triphosphates. That makes the system useful for controlled experiments but different from a cell that can obtain every ingredient from ordinary nutrients.

Translation is a systems problem
Getting an unnatural codon to a ribosome is only one part of protein production. The cell also needs the matching transfer-RNA and aminoacylation arrangements, plus safeguards against losing or misreading the sequence. The 2017 result demonstrated the selected decoding events; it did not eliminate those engineering constraints for all possible sequences.
Biological containment remains important
A semi-synthetic organism that relies on unusual substrates offers a built-in dependency that can help keep experiments controlled, but containment and environmental-release questions still require separate assessment. The reported studies were demonstrations in laboratory systems, not authorization for open release.
How to read the headline accurately
“Bacterium survives unnatural DNA transplant” is a vivid shorthand for a narrower scientific result. The bacterium did not receive a complete foreign genome and then live normally without assistance. Researchers inserted DNA containing an unnatural pair into an engineered strain, supplied the corresponding nucleotide triphosphates, and showed first that the DNA could propagate and later that its information could be retrieved to make a protein.
The most precise one-sentence summary is therefore: an engineered E. coli semi-synthetic system maintained DNA with the dNaM–dTPT3 base pair and, in follow-up work, used that added information to direct amino-acid incorporation in sfGFP.
Evidence timeline
| Year | Paper or finding | Scientific significance |
|---|---|---|
| 2014 | A semi-synthetic organism with an expanded genetic alphabet (Nature) | Established propagation of DNA containing an unnatural base pair in engineered E. coli. |
| 2017 | Zhang and colleagues, A semi-synthetic organism that stores and retrieves increased genetic information (Nature) | Showed transcription, ribosomal decoding and reporter-protein production using the added information. |
Read together, these papers mark a progression from maintaining an expanded DNA sequence to using it as an instruction. That progression—not a naturally occurring bacterial mutation—is the core of the story.
