In 2009, researchers reported that the Alaskan darkling beetle Upis ceramboides contains a highly active antifreeze factor with little or no protein. They described it as a carbohydrate-and-fatty-acid material, but left important structural and biological questions open. A 2024 paper later challenged reports of naturally occurring xylomannans with antifreeze activity, so the original interpretation is not settled.
How does Upis ceramboides survive freezing?
The beetle is a freeze-tolerant darkling beetle collected in interior Alaska. The 2009 paper says it can tolerate freezing to −60 °C in midwinter. That figure describes the beetle’s reported cold tolerance—not the performance of the isolated molecule in a laboratory assay, and it does not by itself establish how the beetle survives.
The study’s striking finding was a thermal-hysteresis-producing factor extracted from adult beetles. Thermal hysteresis is the gap between a solution’s colligative melting point and its lower, hysteretic freezing point. In this context, the effect indicates activity by a large-molecular-mass antifreeze factor. It is distinct from a whole-animal survival test.

Walters and colleagues reported that the isolated material produced 3.7 ± 0.3 °C of thermal hysteresis at 5 mg/ml. They compared its activity with that of the most active insect antifreeze proteins. This result applies to the isolated sample at the stated concentration and under the paper’s assay conditions; it is not a measure of how far below zero the beetle can survive.

The authors characterized their result this way: “This xylomannan is the first TH-producing antifreeze isolated from a freeze-tolerant animal and the first in a new class of highly active THFs that contain little or no protein.” That was the team’s conclusion in 2009, before the later challenge to proposed natural xylomannan antifreeze activity. What structure did the researchers propose?
The 2009 analyses indicated a backbone containing β-mannopyranosyl-(1→4) β-xylopyranose units, along with a fatty-acid component. The authors did not establish a definitive complete structure: they said the sugar sequence was undetermined and might not strictly alternate. They also noted that the lipid might not be covalently attached to the carbohydrate.

Treat these as the original paper’s reported analytical findings and proposed description, not as a settled molecular structure. In the tested sample, treatment with endo-β-(1→4)xylanase eliminated thermal-hysteresis activity. That result links the observed activity to a xylanase-sensitive component in that sample; it does not resolve every structural question or settle the later dispute.

What might the factor do inside the beetle?
The team proposed several possible roles: inhibiting ice recrystallization, preventing extracellular ice from seeding freezing inside cells, or stabilizing cell membranes. These were hypotheses, not demonstrated mechanisms. The 2009 authors explicitly said the factor’s physiological role in the beetle was not well understood, so the assay does not establish that this molecule is what enables whole-animal survival at −60 °C.

Why is the finding disputed?
A 2024 paper in National Science Review, titled “Expeditious chemical synthesis of xylomannans disproves the proposed antifreeze activities,” states that its results disprove previous reports about natural antifreeze xylomannans. Its stated challenge means the 2009 claim should be presented as a historical report, not an uncontested discovery. The available abstract-level record does not establish which precise structural or activity claim was overturned, so a more detailed account of the dispute would require consulting the full paper.
