Texas A&M University researchers have developed a way to image mitochondria inside terminal Schwann cells while those cells remain in their natural muscle environment. The mouse-tissue method, reported on Oct. 12, 2026, revealed different mitochondrial network patterns in healthy and dystrophic tissue, giving scientists a new tool for studying how nerve–muscle connections respond to injury.
What are terminal Schwann cells?
Terminal Schwann cells sit at neuromuscular junctions, the contact points where motor nerves communicate with muscle. They help maintain these connections and participate in repair after an injury.
In an undisturbed junction, a terminal Schwann cell has a distinctive, branching, pretzel-like shape. After nerve damage, it can change shape, move into affected tissue, clear cellular debris, release growth factors and form a pathway that helps a regenerating nerve find its way back to muscle. Cells that become more spherical and mobile are therefore not simply damaged bystanders; their behavior can influence whether a connection is rebuilt.

“If you want to take a look at the things inside of them, as they are in the body, then somehow you have to image them without them knowing anything is disrupted,” said Aaron Morton, an assistant professor at Texas A&M.

Why these cells have been difficult to study
Taking a terminal Schwann cell out of muscle can alter its shape before microscopy begins. That makes it hard to tell whether an observed structure reflects the cell’s biology or the handling procedure.
The surrounding tissue creates a second problem. Muscle contains many mitochondria of its own, so a dye intended to mark mitochondria in Schwann cells can also label nearby muscle. A strong signal may then be impossible to assign confidently to the Schwann cell.

How the new imaging workflow works
The method is described in the Journal of Visualized Experiments article “Methods for In situ Quantification of Mitochondrial Morphology in Muscle and Terminal Schwann Cells of Mice” (DOI 10.3791/69732). Its central strategy is to preserve the junction in place and reduce the time available for the label to spread through surrounding muscle.
- Label early: Researchers label mitochondria inside the terminal Schwann cells before the dye has spread substantially into neighboring muscle.
- Prepare tissue quickly: The muscle is processed rapidly to limit shape changes and other structural disturbance.
- Image in situ: Mitochondrial organization is examined while the Schwann cell remains embedded in its native tissue context.
- Quantify morphology: The resulting images can be used to assess network connectivity, fragmentation and specific configurations within the cell.
This combination addresses both major sources of uncertainty: deformation caused by isolation and signal contamination from muscle mitochondria.

What the microscope method showed
In healthy terminal Schwann cells, mitochondria appeared as interconnected networks. Near branch points, the researchers also saw small, doughnut-shaped loops. In a dystrophic mouse model, the networks were more fragmented and had lost those configurations.

Morton compared the network behavior to social interaction: “When everyone’s happy, they hold hands and exchange contents. When they’re mad, they all break apart and keep to themselves.” The analogy describes the observed organization, not a proven explanation for why the dystrophic pattern occurs.

Why mitochondrial organization matters for nerve repair
Mitochondria are the cell’s energy-producing structures. A terminal Schwann cell that changes shape, migrates, clears debris or releases growth factors may place different demands on its energy system. Seeing whether mitochondria remain connected or become fragmented gives researchers a way to examine the cell’s internal state alongside its role at the neuromuscular junction.
The new workflow does not show that a particular mitochondrial pattern causes successful or failed nerve regeneration. It makes that question testable without first destroying the native geometry that researchers need to study.

Does this prove a treatment for nerve damage?
No. The report describes a laboratory imaging method in mouse tissue, not a therapy, human result or demonstrated improvement in recovery. It provides no numerical accuracy measure and does not establish that changing mitochondrial structure will restore nerve function.
The current study also did not investigate amyotrophic lateral sclerosis, diabetic neuropathy or spinal muscular atrophy. Terminal Schwann cells have been implicated in those conditions in earlier work, so they are possible subjects for later studies rather than diseases addressed by this experiment.
What could come next
Morton’s laboratory plans to apply the technique to nerve-injury experiments and to test whether an experimental biomaterial designed to promote mitochondrial health can improve nerve recovery. That is a planned investigation, not an established treatment or a clinical recommendation.

Because the workflow keeps cell shape and mitochondrial organization visible together, future studies can compare injury stages, disease models and interventions using the same structural measures. Such comparisons will be needed to determine whether fragmentation is a marker of damage, a contributor to it, or both.
