Hard ticks attach in two stages: their mouthparts anchor in the skin, then saliva forms a cement-like cone around the entry point that helps strengthen the hold. A 2024 study found that one glycine-rich protein in blacklegged tick saliva can separate into protein-rich droplets that age toward a gel and show adhesive properties. It offers a clue to tick cement—not a complete explanation of how every tick sticks.
How do ticks stick to your skin?
For hard ticks, attachment begins mechanically. Sensory palps help the tick position itself; chelicerae cut into tissue; and the barbed hypostome penetrates the skin and serves as a channel for blood. Saliva-derived cement then accumulates around the mouthparts and entry site, adding to that initial anchor. A review describes cement cones as characteristic of most hard ticks, though they have not been observed in every Ixodes species.
Hard ticks can remain attached for several days, with duration varying by species and life stage. This prolonged feeding matters because exposure to tick saliva can enable pathogen transmission; the protein study discussed here did not measure transmission or disease.
What is tick cement?
Tick cement is a saliva-derived material that hardens around the mouthparts. It is not simply one protein: reviews describe a mixture that can include hardened glycine-rich proteins, lipids, carbohydrates, and additional protein classes in some species. Its composition and production vary among tick lineages and species, so a finding about one protein should not be treated as a universal recipe.

The scale of that variation is illustrated by a 2019 study of lone star ticks (Amblyomma americanum): researchers identified 67 glycine-rich proteins in saliva, and 60 of those were secreted in abundance within the first four days. Those figures apply to that species and study, not to ticks generally. A 2024 paper also cites earlier work reporting that glycine-rich proteins accounted for 19% of identified cement-cone protein sequences; that is a reported result from prior work, not a measurement made in the new phase-separation experiments.
What did the 2024 protein-cement study find?
The researchers tested one small protein fragment
Ganar and colleagues studied tick-GRP77, a mature, 77-amino-acid fragment from the saliva of Ixodes scapularis, the blacklegged tick. The sequence is approximately 26% glycine and approximately 44% non-polar amino acids, including glycine. The team synthesized the protein and tested it in controlled laboratory solutions. The paper appeared online on 29 November 2024 in Nature Chemistry; it was published in volume 17, pages 186–197, in 2025. Read the study in Nature Chemistry.

Salt and evaporation produced protein-rich condensates
In droplet-evaporation tests, water loss concentrated the protein, and protein-rich condensates formed. Salt also promoted coacervation—a separation into protein-rich and protein-poor regions. Experiments with protein fragments and mutants implicated arginine and aromatic amino-acid residues in this behavior. The authors described cation–π and π–π interactions involving these residues as dominant in GRP77’s phase separation.

The condensates aged and showed adhesion
Over time, the condensates transitioned toward gel-like aggregates. Adhesion measurements found that the condensates were sticky. The researchers also reported protein-rich condensates in natural tick saliva, a finding that supports the relevance of phase separation while not demonstrating that the simplified laboratory system recreates the full process in a feeding tick.
What the protein finding does—and does not—explain
The biological description of cement as saliva that hardens around tick mouthparts predates this study. The new contribution is a proposed physical-chemistry mechanism for how one glycine-rich protein may help create adhesive material: it separates into condensates and changes over time toward a gel-like state. The work does not establish that GRP77 alone makes natural cement, explain every stage of attachment, or show that all ticks use the same mechanism.

The distinction matters because a natural cement cone contains more than the single synthesized fragment tested in solution, and the process occurs in the complex environment of skin and saliva. The study offers a molecular clue, not a complete account of tick attachment.
