A deliberately shortened ribozyme made from three separate RNA strands still catalyzed a chemical reaction in the laboratory. The 2016 study found that this fragmented version worked best at a lower temperature than its full-length parent. It did not revive a dead molecule or demonstrate that life began with RNA; it tested how much catalytic function a small, fragmented RNA assembly could retain.

What the researchers changed

Arvin Akoopie and Ulrich F. Müller took an existing triphosphorylation ribozyme and shortened it by removing its longest double-stranded section and splitting the RNA into separate strands. The shortest construct comprised a 14-nucleotide substrate and two ribozyme fragments, 34 and 19 nucleotides long. Even with this reduced structure, the construct retained catalytic activity. The primary paper was first published by the Royal Society of Chemistry on 7 April 2016.

What reaction did the ribozyme catalyze?

A ribozyme is RNA that acts as a catalyst. This one catalyzed a reaction between an RNA molecule’s 5′-hydroxyl group and trimetaphosphate, adding a 5′-triphosphate to the RNA. The study was about triphosphorylation—not RNA replication, genome repair, or the creation of a living system.

The researchers discussed the reaction as a possible way for a prebiotic RNA-based system to access chemical energy. That makes it relevant to the RNA-world hypothesis, which considers whether RNA could have played a central role in early life. It is a model reaction, not evidence that RNA-only organisms existed or that this particular ribozyme occurred on early Earth.

How did fragmentation affect performance?

The key result was a change in temperature response, not a universal improvement in speed. The shortened fragmented construct had an optimum around 20 °C; the paper reports a range of 15–25 °C. Its full-length parent had an optimum around 40 °C, according to the Royal Society of Chemistry’s report of the study.

At 22 °C, the shortest fragmented construct’s reaction kinetics were about two-fold lower than those of the full-length ribozyme. Some intermediate constructs, however, had faster kinetics than the parent under the tested conditions. The comparison therefore depends on both the construct and the assay temperature; it would be misleading to say simply that fragmentation made the ribozyme better.

What the result suggests—and what it does not

The experiment shows that a ribozyme can retain catalytic activity after substantial shortening and division into small RNA strands. Its lower temperature optimum suggests that cooler conditions may favor activity in some fragmented constructs. The authors proposed that lower temperatures might stabilize ribozymes assembled from short RNA fragments.

This is a laboratory result that informs possible scenarios for prebiotic chemistry. It does not establish that the tested RNA assembly formed naturally, that early Earth had this exact chemistry, or that the RNA-world hypothesis is proven. The Chemistry World report noted a Sun about 25% dimmer in the early-Earth context; that figure is background, not a measurement from the ribozyme experiment.

How this differs from a later RNA-repair study

A separate study reported by the University of Notre Dame on 13 July 2026 concerns an engineered ribozyme that joins broken RNA and recognizes terminal phosphate groups. That is a different ribozyme and a different catalytic function from the 2016 triphosphorylation experiment. The Notre Dame report says researchers are still working to improve reaction efficiency and broaden the range of targets, so possible diagnostic relevance remains a potential application, not an established clinical product.
