In a 2021 study, researchers chemically made a mirror-image version of the DNA-copying enzyme Pfu DNA polymerase and used it to assemble a 1.5-kilobase mirror-image 16S rRNA gene. The result was a major laboratory demonstration—not a living mirror organism, a functional mirror ribosome, or proof that 1.5 kb remains the longest mirror-DNA construct ever reported.
What is mirror-image DNA?
DNA’s familiar double helix is built from sugar molecules with a particular handedness, called D-deoxyribose. In L-DNA, the sugar has the opposite handedness. The two forms are mirror images, and ordinary biological enzymes generally interact with them differently. That difference makes L-DNA interesting for research: it can resist degradation by enzymes such as natural DNase I that break down ordinary DNA.
Mirror-image DNA is not simply ordinary DNA twisted the other way. Making and copying it requires compatible mirror-image molecular tools. In the 2021 work, Chuyao Fan, Qiang Deng and Ting F. Zhu chemically synthesized a mirror-image form of Pfu DNA polymerase, an enzyme from Pyrococcus furiosus. The primary study, published in Nature Biotechnology, describes a roughly 90-kDa enzyme based on a 775-amino-acid natural polymerase.
How did the researchers make the 1.5-kb construct?
Building a large enzyme from chemically synthesized pieces is challenging. The team used a split-protein design and amino-acid substitutions to make chemical synthesis of the mirror-image Pfu polymerase feasible. With that enzyme, they assembled a mirror-image 16S rRNA gene from three blocks.

The assembled gene was 1.5 kilobases long. It was a DNA sequence intended as a template for future transcription, not a working ribosome. The authors presented mirror-image translation and a functional mirror-image ribosome as future steps, rather than results of this experiment.
What did “longest ever” mean?
The “longest ever” wording refers to the achievement reported in 2021: a 1.5-kb mirror-image gene assembled with a chemically synthesized mirror-image polymerase. It should be treated as a dated claim, not a confirmed current record. The later studies cited here extended L-DNA research, but do not establish that a longer mirror-image DNA strand has—or has not—since been reported.

A 2022 follow-up used mirror-image polymerase tools to select biostable L-DNA aptamers against native human thrombin, expanding the demonstrated applications beyond information storage. In 2025, a separate study reported a one-pot synthesis method for mirror-image Dpo4, a different polymerase. Its reported overall yield was 9.8%, described by the authors as about twice the prior yield; that is a synthesis-efficiency result, not a longer-DNA record.

In the 2021 study, the synthetic L-polymerase had a measured error rate of 3.6 × 10−6 on a 1.2-kb template. After the researchers improved oligonucleotide purification, about 90% of final assembled sequences were reported correct; the remaining sequences had single-nucleotide substitutions. These are results from defined laboratory experiments, not a guarantee of the same fidelity for every template or process.

What did the L-DNA information-storage test show?
The researchers encoded a paragraph from Louis Pasteur’s 1860 writing in L-DNA segments, then demonstrated amplification and recovery through sequencing. In a pond-water experiment, a trace L-DNA barcode remained amplifiable and sequenceable after one year. Under the same reported conditions, a D-DNA barcode could not be amplified after one day. This is a specific experimental comparison, not a general shelf-life or environmental-survival guarantee.
The result illustrates the idea of bioorthogonal storage: L-DNA can be handled separately from many natural DNA-based biological processes because ordinary biological degradation mechanisms do not act on it in the same way. It does not make L-DNA indestructible. The 2021 paper notes susceptibility to physical stress and chemical degradation, alongside practical obstacles including high cost, slow writing and reading, inconvenient sequencing, and a lack of error-correction mechanisms.

What the work does—and does not—establish
- Established: the team chemically synthesized a roughly 90-kDa mirror-image Pfu DNA polymerase and used it to assemble a 1.5-kb mirror-image 16S rRNA gene.
- Also demonstrated: laboratory copying and sequencing-based retrieval of information encoded in L-DNA, plus a specific pond-water persistence comparison.
- Not demonstrated: a functional mirror-image ribosome, a mirror-image organism, or a commercial DNA-storage system.
- Not established by the cited later work: whether the 1.5-kb construct remains the longest mirror-image DNA strand reported.
