The 2019 method from Mahesh Vishe and Jeffrey N. Johnston builds stereocontrolled β2,3-amino amides by reversing polarity twice: an enantioselective nitroalkane addition to a β-substituted nitroalkene makes the key carbon–carbon bond, then Umpolung Amide Synthesis (UmAS) installs the amide without first making a carboxylic acid or activated acid derivative. A molybdenum oxide-mediated hydride reduction is reported to deliver the anti-β2,3-amino amide with high selectivity under the authors’ standard conditions.
What the paper actually demonstrates
Vishe and Johnston reported a stereocontrolled synthesis of β2,3-amino amides in Chemical Science in 2019. The article, titled The inverted ketene synthon: a double umpolung approach to enantioselective β2,3-amino amide synthesis, appeared in volume 10, pages 1138–1143; it was first published online on 12 November 2018 and is open access. The DOI is 10.1039/C8SC04330B.

The target class matters because β-amino amide motifs combine an amide functional group with a nearby stereochemically defined amino-bearing carbon framework. Such motifs occur in complex natural products and therapeutic molecules, but constructing a chosen relative and absolute configuration can be difficult. A 2018 Chemistry World report placed the work in that broader synthetic context; it should be read as background rather than as evidence of a clinical or quantitative advantage.
This is a proof of a particular synthetic strategy, not a claim that every β-amino amide substrate will react efficiently or that the route is universally better than established alternatives. The accessible publication material does not provide a complete, independently verifiable substrate-by-substrate table of yields and enantiomeric ratios, so those values should be taken from the full article and supporting information before being quoted.

What “double umpolung” means in this synthesis
Umpolung means deliberately reversing the normal polarity of a synthetic building block. In a conventional amide disconnection, an acyl component behaves as an electrophile and an amine behaves as a nucleophile. The Johnston strategy changes that logic in two distinct bond-forming events.
First polarity reversal: making the carbon framework
A nitroalkane is used as the carbon nucleophile and adds to a β-substituted nitroalkene. The resulting carbon–carbon bond is therefore made through a polarity pattern that differs from the usual way chemists assemble an acyl-derived fragment. This enantioselective addition establishes the carbon skeleton and creates the stereochemical information needed for the β2,3 relationship.
Second polarity reversal: making the amide
The next operation is Umpolung Amide Synthesis, abbreviated UmAS by the authors. Instead of converting an alcohol or other precursor into a carboxylic acid and then activating that acid for amine coupling, UmAS forms the amide directly from a different polarity arrangement. The route therefore reaches the amide without an intervening carboxylic acid or activated acid derivative.
The phrase double umpolung describes the combination of these two choices: the carbon–carbon bond is formed through the inverted nitroalkane/nitroalkene polarity pattern, and the carbon–nitrogen amide bond is formed by UmAS rather than by ordinary acyl activation.

How the sequence is organized
- Select the nitroalkene partner. The key alkene is β-substituted so that addition can encode the substitution pattern and stereochemistry of the eventual β2,3-amino amide.
- Perform the enantioselective nitroalkane addition. Nitroalkane addition creates the central carbon–carbon connection. The stereochemical outcome at this stage is carried into the downstream product rather than being introduced only at the final amide coupling.
- Apply the reduction and amide-forming operations. In the reported sequence, a molybdenum oxide-mediated hydride reduction is associated with formation of the anti-β2,3-amino amide, while UmAS supplies the direct amide construction. The available summary does not justify assigning a more detailed reagent-by-reagent mechanism than the authors report.
- Characterize stereochemical outcome. The authors measured enantiomeric ratios by high-performance liquid chromatography using a chiral stationary phase. Anti describes the relative relationship of the relevant substituents; it is not, by itself, a full absolute-configuration assignment.
This order is strategically important. The carbon framework and its stereochemical bias are established before the amide is installed, so the amide step does not have to solve the entire stereochemical problem at once.
Reported standard conditions
The scheme caption gives a concise set of standard operating conditions. They are the authors’ reported conditions, not an independent replication or a guarantee of performance on a new substrate.
| Parameter | Reported value | How to interpret it |
|---|---|---|
| Reaction scale | 0.1 mmol | A small-scale demonstration; scale-up behavior is not established by this number. |
| Concentration | 0.1 M | The stated standard concentration for the reactions shown. |
| Amine loading | 2 equivalents | Applies to the standard reactions described in the scheme caption. |
| Temperature | 25 °C | Ambient-temperature operation as reported. |
| Reaction time | 24 hours | The stated standard hold time, not a universal completion time. |
| Enantiomeric-ratio measurement | Chiral-stationary-phase HPLC | The analytical method used to assess enantioselectivity. |
The accessible summary does not expose a complete numerical yield and enantiomeric-ratio table. It is therefore inappropriate to replace the authors’ description of “high selectivity” with an invented percentage or to treat one substrate’s result as a general value.
Why the anti product is significant
β2,3-amino amides contain more than one stereochemical relationship in a compact part of the molecule. The anti product has a defined relative arrangement between the relevant substituents on adjacent stereogenic positions. Obtaining that relationship selectively reduces the need to separate large amounts of diastereomeric material later.
Relative stereochemistry and enantiomeric purity answer different questions. A reaction can favor the anti diastereomer while still producing both enantiomers, or it can be highly enantioselective without giving the desired anti relationship. The paper addresses both aspects through its stereocontrolled design and chiral-HPLC analysis, but the summary available here does not provide numerical values that can be generalized across the substrate set.
What the “inverted ketene synthon” idea contributes
The article’s title calls the strategy an inverted ketene synthon approach. The useful design insight is retrosynthetic: a chemist can view the target amide as if it arose from a ketene-like acyl fragment, but then construct that fragment through reversed polarity rather than generating a conventional ketene or activated carboxylic acid. This viewpoint makes the nitroalkene/nitroalkane adduct a precursor to the amide skeleton instead of treating it merely as a protected amino-alcohol equivalent.
That change can simplify the logic of a synthesis. The carbon–carbon bond is made where the target needs it, and the amide is formed directly from the resulting polarity pattern. Fewer functional-group interconversions can be required than in a route that first builds a carboxylic acid, activates it, and then couples an amine. Whether it is actually shorter or more efficient for a particular molecule depends on substrate behavior, purification, protecting-group needs and the full experimental procedure.
A practical reading of the method
A chemist planning to apply this chemistry should treat the published scheme as a starting point rather than as a complete transfer protocol. Before attempting a new substrate, obtain the full experimental section and supporting information and verify the identity, preparation and purity requirements for every nitroalkene, nitroalkane, amine, hydride source and molybdenum reagent.

Minimum setup checklist
- Confirm the substitution pattern of the nitroalkene and how it maps onto the desired β2,3 stereochemistry.
- Use the stated 0.1 mmol scale and 0.1 M concentration for an initial test unless the complete procedure specifies otherwise.
- Charge the amine at the reported 2-equivalent loading for a standard reaction.
- Hold the reaction at 25 °C for the stated 24-hour period, while monitoring for substrate consumption and side products.
- Record the exact order of addition, atmosphere, solvent, catalyst loading and workup from the full procedure; those details are not supplied in the summary conditions.
- Analyze both diastereomeric relationship and enantiomeric ratio. Chiral HPLC conditions must be established for the specific product rather than assumed from another substrate.
What not to infer from a successful small-scale run
- A 0.1 mmol example does not establish safe or economical operation at gram scale.
- High selectivity for the reported examples does not prove broad compatibility with electron-rich, electron-poor, sterically hindered or heterocyclic partners.
- The absence of an acid intermediate in this route does not mean that every amide substrate can be made without protecting-group or purification challenges.
- A favorable anti/ syn ratio is not the same as a high isolated yield or a high enantiomeric ratio.
How this route differs from a conventional amide synthesis
| Design question | Conventional acyl-coupling logic | Double-umpolung logic in the 2019 paper |
|---|---|---|
| Key carbon–carbon bond | Often made through a standard nucleophile/electrophile pairing selected around an acyl precursor. | Made by enantioselective nitroalkane addition to a β-substituted nitroalkene. |
| Amide formation | Carboxylic acid is commonly converted to an activated derivative before amine coupling. | UmAS forms the amide without an intervening carboxylic acid or activated acid derivative. |
| Stereochemical plan | May require asymmetric coupling, chiral starting material, resolution or downstream separation. | Stereochemical information is introduced during the nitroalkane addition and carried into the amide product. |
| Evidence level | Depends on the specific established reaction and substrate. | Demonstrated as a 2019 method for the reported examples; universal scope is not established. |
This comparison describes bond-disconnection logic, not a claim that one family of reactions always uses fewer steps, gives higher yield or is safer. Those conclusions require a substrate-specific comparison using complete procedures and data.

Calling every β-amino amide a β2,3-amino amide
The β2,3 designation refers to a particular substitution and stereochemical framework. A generic molecule containing an amino group at a beta position may not have the same connectivity or relative configuration as the paper’s targets.
Equating “anti” with a single absolute configuration
Anti is a relative descriptor. Absolute configuration requires structural assignment and comparison with the authors’ nomenclature or analytical data.

Assuming UmAS is ordinary peptide coupling
UmAS is the defining polarity-reversed amide-forming operation. Describing the method as a routine acid activation and coupling erases the central innovation.
Best Value
Turning qualitative language into a number
The paper’s summary describes the molybdenum oxide-mediated hydride reduction as giving the anti product with high selectivity. No verified numerical selectivity should be supplied unless it is read directly from the complete article or supporting information.
Limits of the current evidence
The article establishes the concept and reports standard conditions, but the material summarized here does not independently verify every substrate entry, isolated yield, absolute configuration, catalyst loading or operational detail. It also does not establish scale-up, long-term reagent stability, broad functional-group tolerance or a head-to-head advantage over all other β-amino amide syntheses.
Those limits do not weaken the central result. They define how it should be used: as a documented 2019 strategy for stereocontrolled β2,3-amino amide construction, with the full paper and supporting information consulted before selecting a substrate, quoting a number or planning production-scale work.

Vishe and Johnston’s double-umpolung approach solves two connectivity problems with reversed polarity. Enantioselective nitroalkane addition to a β-substituted nitroalkene builds the carbon framework, and UmAS installs the amide without an acid or activated-acid intermediate. Under the reported 0.1 mmol, 0.1 M, 25 °C, 24-hour conditions with 2 equivalents of amine, molybdenum oxide-mediated hydride reduction is described as delivering the anti β2,3-amino amide with high selectivity. It is a significant, specific method—not a universal recipe—and numerical scope claims should come from the complete publication record.
