A 2015 study combined atomic force microscopy (AFM) and scanning tunnelling microscopy (STM) to examine more than 150 individual asphaltene molecules. Among the molecules characterized, the most common architecture was an aromatic “island”—one fused aromatic core surrounded by alkyl side chains. A smaller set showed separate aromatic regions connected by chains or single bonds, consistent with an “archipelago” arrangement.

The result was an important direct-imaging observation, not a claim that every asphaltene molecule has one universal structure. Asphaltenes are a complex mixture, and the study’s value was showing molecular architectures that bulk measurements often blend together.

What are asphaltenes?

Asphaltenes are a complex fraction of crude oil made up of many hydrocarbon molecules rather than one pure chemical compound. Their structures can include polycyclic aromatic hydrocarbon regions, alkyl side chains and other variations in size, shape and connectivity. Because the mixture contains many related species, describing it with one molecular formula or one representative drawing is misleading.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

The molecules also tend to associate with one another in solution. When many species associate and are measured together, an analytical signal represents an ensemble rather than an individual molecule. That makes it difficult to determine which structural features belong to which molecules. The 2015 work addressed this problem by separating molecules onto a surface and imaging them one at a time.

This distinction between a fraction and a single compound is important for interpreting every claim about asphaltene “structure.” A sample can contain molecules with different aromatic sizes, different numbers of side chains and different ways of connecting those parts. A description that fits one molecule may therefore be useful without serving as a formula for the whole petroleum fraction.

Why individual-molecule imaging mattered

Before this work, researchers had evidence for different structural descriptions of asphaltenes, but conventional measurements generally averaged signals from large populations. An average can show that aromatic groups, alkyl groups or particular chemical bonds are present without revealing how those groups are arranged within each molecule.

Two arrangements were especially important in the debate:

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.
  • Island model: aromatic rings are fused into one dominant central core, with alkyl chains attached around its perimeter.
  • Archipelago model: two or more aromatic regions remain separate and are connected by alkyl chains or, in the structures described in the report, by single bonds.

These labels are useful models, not a rule that divides every asphaltene into one of two permanent categories. A natural sample can contain molecules with different architectures, and a molecule’s observed appearance depends on the sample and measurement conditions.

The value of seeing individual molecules is therefore statistical as well as visual. Once molecules are examined separately, recurring patterns can be distinguished from occasional ones, and unlike structures need not be hidden inside one averaged spectrum or image. The method does not remove the mixture’s complexity; it makes that complexity observable.

How the 2015 AFM and STM experiment worked

The researchers used a specialized surface-science preparation rather than attempting to image molecules inside liquid crude oil. A solid asphaltene sample was heated in an ultrahigh-vacuum environment under low-temperature conditions. Individual molecules detached from the solid and landed on a substrate, where the instruments could examine them separately.

  1. Prepare a solid sample. The asphaltene material was placed in an ultrahigh-vacuum instrument so that the surface would not be obscured by air, solvent or contaminants.
  2. Release individual molecules. Heating caused molecules to detach from the solid. They traveled through the vacuum and deposited on a substrate maintained under low-temperature conditions.
  3. Image the surface with AFM. The atomic force microscope traced the surface topography of deposited molecules. In this experiment, AFM supplied information about the molecules’ atomic structure and physical outline.
  4. Measure with STM. Scanning tunnelling microscopy added information related to the shape of the molecules’ electronic or molecular orbitals. That information complemented the physical height and structure seen with AFM.
  5. Compare many molecules. The team characterized more than 150 individual molecules. The count describes the analyzed set reported in 2015; it is not an estimate of the proportion of island or archipelago molecules in all crude oil.

Working in vacuum and on a substrate is essential to this type of experiment. It provides a stable, clean surface for high-resolution measurements, but it also means the result should be interpreted as an observation of deposited molecules under controlled laboratory conditions, not a movie of asphaltenes behaving in an operating reservoir or refinery.

The preparation also explains why the experiment is different from simply putting crude oil under an ordinary microscope. The molecules had to be released from a solid, isolated on a suitable surface and held in conditions that allowed the probe and tunnelling junction to operate. Those steps are what make individual-molecule resolution possible, while also defining the boundaries of the result.

What AFM contributed, and what STM added

Method Information supplied in the study Why it was useful
Atomic force microscopy (AFM) Surface topography and atomic-scale structural detail Revealed the physical arrangement and outline of individual molecules on the substrate
Scanning tunnelling microscopy (STM) Information about molecular-orbital shape Added electronic-structure information that helped interpret features not defined by height alone

Neither instrument by itself turns a complex petroleum fraction into a single definitive formula. AFM and STM are complementary: AFM is primarily a structural image, while STM contributes information about electronic states and orbital shape. Considering both measurements helps distinguish a real molecular feature from an imaging artifact or a feature that merely appears because of the molecule’s position on the surface.

That complementarity matters especially for aromatic systems. A raised feature in a topographic image is not automatically a separate aromatic ring, and an electronic contrast in an STM image is not simply a height measurement. Interpreting the two kinds of information together gives a more defensible picture of how aromatic regions and side chains are arranged.

Island and archipelago models compared

Feature Island arrangement Archipelago arrangement
Aromatic rings Fused into one main aromatic core Present in two or more aromatic regions
Connections between regions No separate aromatic islands are required; the rings share fused edges Regions can be linked by alkyl chains or by single bonds
Alkyl groups Typically form peripheral chains around the core Can serve as bridges or occur as substituents on separate aromatic regions
Status in the 2015 observations Dominant arrangement among the molecules characterized Seen in some of the molecules characterized

The distinction is about connectivity, not simply about whether a molecule contains aromatic rings. Both arrangements can contain aromatic and aliphatic portions. The question is whether most of the aromatic rings belong to one fused core or to separate aromatic units that remain connected by other bonds or chains.

“Island” also does not mean that a molecule is isolated from every other molecule in the oil. It describes the internal architecture of one molecule: a concentrated aromatic region with alkyl material around it. “Archipelago” similarly describes how aromatic regions are connected within a molecule, not whether molecules form aggregates in solution.

What the study reported finding

The central observation was that most of the more than 150 molecules examined had the island architecture: a central aromatic region with alkyl chains around it. The researchers also observed molecules with aromatic regions connected in a way compatible with the archipelago model.

The report did not provide a population-wide percentage, and the number of molecules examined should not be converted into a claim about the fraction of all asphaltenes in every crude oil. “Dominant” describes the distribution within the reported set of deposited molecules. Different crude-oil sources, preparation methods or molecular populations could produce different distributions.

The finding therefore resolves part of a structural question without eliminating molecular diversity. It supports the view that a single asphaltene sample can contain several architectures, while showing that the island form was more frequently observed in this particular study.

More than 150 is substantial for direct molecular inspection, but it remains a defined experimental set, not a census of the world’s crude oils. The study’s strength is the clarity of the observations and the ability to compare molecules individually, not a claim that the counted set establishes a universal prevalence.

Why bulk measurements had struggled to settle the question

Ensemble methods combine signals from many molecules. If one molecule has a large fused aromatic core and another has two smaller aromatic regions joined by a chain, their combined signal can indicate aromatic content without clearly identifying the connectivity in either molecule. Association in solution adds another complication: molecules can cluster, changing the measured response compared with isolated species.

Direct imaging changes the question from “What average signal does the mixture produce?” to “What does this individual deposited molecule look like?” Examining many individual molecules then allows researchers to see whether one architecture recurs, whether alternatives coexist and how much structural variation is present. The approach does not replace bulk chemical analysis; it supplies a level of structural detail that bulk data alone may not provide.

This is why the 2015 result is best understood as a bridge between molecular pictures and mixture-level measurements. Bulk methods remain necessary for describing the whole asphaltene fraction, while AFM and STM show how structures contributing to that fraction can be organized one molecule at a time.

What the result means for crude-oil properties

Asphaltenes affect properties relevant to petroleum production, transport and refining. Their tendency to associate, deposit or alter fluid behavior depends on molecular interactions as well as composition. A more realistic account of molecular size, aromatic connectivity and side-chain arrangement could eventually improve models that describe those behaviors.

That implication remains prospective. The imaging experiment itself did not demonstrate improved reservoir performance, easier transport, better refining yields or enhanced oil recovery. Chemistry World quoted IBM Research’s Leo Gross describing the work as a possible foundation for improving upstream and downstream production and enhanced oil recovery “from first principles” through better molecular models. In context, that is a proposed application of the structural knowledge, not a measured operational result.

The same report quoted Douglas Kushnerick of ExxonMobil Research and Engineering, who called the direct observation unusually clear and suggested that efficiently identifying structures could help address industrial chemistry problems. That comment likewise describes potential future use rather than a validated process change.

A molecular model informed by observed architectures could, in principle, represent mixtures more realistically than a model built around one idealized drawing. It might help researchers connect aromatic connectivity and side-chain arrangement with association or deposition tendencies. But making that connection requires further measurements and modelling beyond the imaging experiment described here.

How to interpret the “first step” language

The phrase “first step” refers to the method’s role in making individual asphaltene structures visible, not to a complete solution to petroleum chemistry. Imaging more than 150 molecules is a substantial advance over seeing only an ensemble average, but it does not identify every compound in a crude oil or establish a universal asphaltene blueprint.

Several layers of interpretation should remain separate:

  • Individual-molecule result: the study observed recurring island-like structures and some archipelago-like structures in its analyzed set.
  • Sample composition: the result describes the asphaltene material used for the experiment, not all crude-oil sources.
  • Bulk behavior: transport, deposition and refining behavior depend on mixtures and interactions, not on one isolated image.
  • Industrial application: improved modeling and production were suggested as longer-term possibilities, not demonstrated outcomes of the experiment.

Keeping these layers separate prevents two opposite mistakes: treating a striking image as a complete chemical inventory, or dismissing it because a surface experiment does not reproduce a reservoir. Its contribution is narrower and more concrete—direct evidence that different molecular connectivities can be observed in a complex asphaltene sample.

What the experiment did not establish

  • It did not prove that every asphaltene molecule is an island.
  • It did not show that archipelago structures are absent; some were observed.
  • It did not supply a universal percentage for the two arrangements.
  • It did not measure all molecular sizes, weights or architectures across every crude oil.
  • It did not reproduce the behavior of asphaltenes in a flowing reservoir, pipeline or refinery unit.
  • It did not by itself prove improved oil recovery or a change in industrial operations.

These limits do not weaken the direct-imaging result. They define what can legitimately be concluded from it. In particular, the terms “most” and “dominant” must stay tied to the molecules characterized in the 2015 report.

Why preparing molecules on a substrate changes the interpretation

Depositing molecules on a solid surface makes high-resolution AFM and STM possible, but the surface is part of the experimental context. A molecule can adopt a particular orientation after landing, and the measured image represents that deposited state under vacuum and low-temperature conditions. The procedure is designed to reveal architecture, not to reproduce the full chemical environment of crude oil.

For that reason, the strongest wording is “the study reported” or “among the molecules characterized.” It is more precise than saying that asphaltenes in general have one fixed shape. The distinction matters whenever a result from a carefully prepared laboratory sample is applied to a chemically diverse natural mixture.

The substrate-based approach also explains why the article should not describe the experiment as imaging liquid crude oil directly. Molecules were detached from a solid sample and inspected after deposition. The experiment captured stable, high-resolution views under controlled conditions; it did not follow association, flow or deposition in real time inside an industrial system.

How the imaging answers common reader questions

What are asphaltenes made of?

They are a complex mixture of hydrocarbon molecules containing aromatic regions and aliphatic, often alkyl, portions. The mixture includes structural variants rather than one compound with one fixed arrangement.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Did AFM reveal a single asphaltene structure?

No. AFM and STM revealed a distribution of individual structures. The island arrangement was dominant in the reported set, while some molecules were compatible with the archipelago arrangement.

Are island and archipelago molecules mutually exclusive categories for all samples?

No. They are structural models used to describe different connectivities, and one sample can contain both types or structures that do not fit a simple label perfectly.

Why was STM needed if AFM already showed the molecules?

AFM supplied atomic-scale structural information, while STM added information about molecular-orbital shape. The two measurements provide different, complementary evidence.

How can researchers image individual molecules if asphaltenes associate in solution?

The reported procedure avoided imaging the associated liquid mixture. A solid sample was heated in ultrahigh vacuum, molecules detached and landed separately on a substrate held under low-temperature conditions, and AFM and STM then examined those deposited molecules.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Does “dominant” mean that almost all asphaltenes are islands?

No. The report gave no universal percentage. “Dominant” refers to the distribution among the more than 150 molecules characterized in that study, not to every asphaltene molecule in every crude oil.

Can the images be used to predict better oil recovery?

They may contribute to future molecular models, but the 2015 experiment did not test or prove an improvement in oil recovery. Any production benefit remains a proposed application.

What is the practical difference between an island and an archipelago?

An island has aromatic rings fused into one main core, with alkyl chains around it. An archipelago has separate aromatic regions connected by alkyl chains or single bonds. Both can occur in the same broad asphaltene fraction.

People and publication behind the report

Chemistry World’s 11 August 2015 report, written by Simon Hadlington, described the work and quoted members of the research team. Bruno Schuler of IBM Research in Zurich said that basic properties such as the size, weight and architecture of asphaltene molecules had been debated for years. The report also quoted Leo Gross, Schuler’s IBM Research co-worker, on possible upstream, downstream and enhanced-oil-recovery applications.

The report quoted Schuler saying, “Current knowledge about this material is quite limited and even some of the basic properties of the molecules in asphaltenes, such as their size, weight and architecture, have been under debate for years.” Gross said that the fundamental study could help improve production and enhanced oil recovery “from first principles,” meaning from molecular models rather than only empirical fits.

Bottom line for readers

The 2015 AFM-and-STM study made a difficult petroleum-chemistry problem more concrete by imaging individual asphaltene molecules. In the reported sample, a single fused aromatic core with surrounding alkyl chains—the island model—was the dominant architecture, but archipelago-like molecules were also present. The advance was direct structural evidence from individual molecules; it was not a universal formula for asphaltenes or proof of an immediate industrial improvement.

Frequently Asked Questions

What did the 2015 AFM study reveal about asphaltenes?

Among more than 150 molecules characterized, the study reported that island-like molecules—with one fused aromatic core and peripheral alkyl chains—were dominant. Some molecules had separate aromatic regions connected by chains or single bonds, consistent with the archipelago model.

Special offer. See more information about Outbyte and uninstall instructions. Please review EULA and Privacy policy.

Were all the imaged molecules the same?

No. The observations showed molecular diversity. The island arrangement was more common in the reported set, but archipelago-like structures were also seen, and no universal percentage was given.

Were asphaltenes imaged directly in crude oil?

No. Solid asphaltene was heated in ultrahigh vacuum, individual molecules detached and deposited on a low-temperature substrate, and the deposited molecules were then examined with AFM and STM.

What is the difference between AFM and STM in this experiment?

AFM supplied information about atomic structure and surface topography. STM added information about molecular-orbital shape and electronic structure. Their measurements were complementary.

Does the finding apply to every crude oil?

Not automatically. The result describes the molecules characterized in the 2015 study. Asphaltene composition varies, so the observed distribution should not be treated as a population-wide rule.

Did the study prove improved enhanced oil recovery?

No. IBM Research’s Leo Gross described improved upstream, downstream and enhanced-oil-recovery modelling as a possible long-term application. The imaging experiment itself did not measure an operational improvement.

The Bottom Line

The 2015 AFM-and-STM work directly imaged more than 150 asphaltene molecules and found an island-dominant pattern in that sample, alongside archipelago-like structures. It clarified molecular diversity without creating a universal asphaltene formula or proving an immediate production benefit.
