Microcrystal electron diffraction (MicroED) can determine a small molecule’s crystal structure from crystals far too small for conventional single-crystal X-ray diffraction. In a 2018 study, researchers collected atomic-resolution electron-diffraction data from nanocrystals of about 100 nanometres, obtaining structures at better than 1 Å resolution in minutes under the study’s experimental conditions.
That is the useful meaning of “certainty” in the original Chemistry World headline: diffraction supplied an unambiguous crystallographic structure for the demonstrated compounds. It is not a guarantee that every molecule, powder or laboratory will produce a structure, and it does not make NMR, X-ray crystallography or other chemical analyses obsolete.
What MicroED actually measures
MicroED is a cryo-electron microscopy method that records electron diffraction from a microcrystal and uses the resulting reflections to solve its crystal structure. Electrons interact strongly with matter, so a crystal only tens to hundreds of nanometres across can produce measurable diffraction. That scale is the central advantage when a compound forms only very small crystallites or a powder that is difficult to grow into an X-ray-sized single crystal.

The output is a crystallographic model: the periodic arrangement of atoms in the crystal, including molecular connectivity and three-dimensional geometry when the data support it. Diffraction is direct structural evidence from the ordered lattice. Spectroscopy, by contrast, often identifies functional groups, environments or connectivity indirectly through signals that must be interpreted together. This distinction does not make spectroscopy unreliable; it explains why MicroED can answer a different question when a direct crystal structure is needed.

Why electrons make tiny crystals useful
X-ray diffraction generally benefits from a relatively large, well-formed single crystal. MicroED uses electrons in a transmission electron microscope, where nanocrystals can be selected and tilted while diffraction is recorded. The technique therefore targets a practical gap: material that is crystalline enough to diffract but unavailable as a conventional X-ray single crystal.
Samples are commonly examined at cryogenic temperature to reduce radiation damage and preserve the specimen in the microscope vacuum. “Cryo” describes the specimen environment; it does not mean that every cryo-EM experiment is an imaging experiment. MicroED is primarily a diffraction measurement.
How the 2018 MicroED workflow determines a structure
The Jones and co-authors study in ACS Central Science (2018) demonstrated a workflow built around ordinary powders and very small crystals rather than elaborate crystal growth. The exact operating parameters depend on the microscope, detector and compound, but the logic is consistent:
- Obtain crystalline material. The starting powder must contain crystallites that are sufficiently ordered to produce interpretable diffraction. MicroED cannot recover a crystal structure from a completely amorphous sample.
- Disperse a small amount on an electron-microscopy grid. The grid supports particles that can be located and examined in the transmission electron microscope. Preparation can be minimal compared with growing a large single crystal, but the sample still has to be compatible with grid handling and vacuum transfer.
- Locate a suitable crystallite. The operator identifies a particle with useful morphology and diffraction quality. A powder can contain multiple phases, so selecting and checking individual crystallites matters.
- Record diffraction while changing orientation. Electrons diffract from the lattice as the crystallite is tilted or rotated. The series of patterns provides reciprocal-space coverage for structure solution.
- Process intensities and solve the crystal structure. Software indexes the reflections, estimates intensities and applies crystallographic methods to produce a model. Refinement and inspection are required to distinguish a valid solution from an artifact or an incorrect phase.
In the reported examples, data collection took minutes and structures were obtained at better than 1 Å resolution. Those are measured outcomes for the tested materials and instrument setup, not service-level promises for all MicroED experiments.

What “organic analysis certainty” means—and what it does not
The headline’s certainty refers to the ability of diffraction data to settle the tested compounds’ crystal structures without relying solely on inferred connectivity. A successful model can distinguish constitutional isomers, reveal stereochemical arrangement when the diffraction data support it, and expose details that may be ambiguous in a single spectroscopic measurement.
Several qualifications are essential:
- A crystal structure describes the ordered solid that was measured. It does not by itself prove bulk purity, quantify impurities, establish solution behavior or predict biological activity.
- Many molecules do not crystallize readily, and some crystals are too disordered, too small or too radiation-sensitive for a usable dataset.
- Polymorphs, solvates and mixtures can produce different diffraction patterns. A structure from one crystallite may not represent every particle in a batch.
- Structure determination still requires crystallographic expertise, suitable software and independent checks of the model.
Consequently, MicroED is best viewed as an additional direct-structure tool. It can complement NMR, mass spectrometry, infrared or Raman spectroscopy, chromatography and conventional X-ray diffraction rather than automatically replacing them.
What the 2018 study demonstrated
Chemistry World’s 2018 coverage of the Jones study reported tests on 12 different compounds, a four-powder mixture and crushed ibuprofen tablets. The examples were important because they showed that the method could work with practical powder samples rather than only ideal laboratory crystals.
| Reported result | How to interpret it |
|---|---|
| Crystallites around 100 nm | Nanocrystals in this approximate size range yielded usable diffraction in the demonstrated experiments. |
| Better than 1 Å resolution | The reported datasets supported atomic-scale structural detail for the tested compounds. |
| Data collection in minutes | Electron-diffraction acquisition was rapid under the study’s conditions; total preparation, screening, processing and refinement can take longer. |
| 12 compounds, a four-powder mixture and crushed tablets | The Chemistry World account used these examples to illustrate performance on multiple compounds and real powdered material. |
A mixture is a particularly useful demonstration because diffraction can reveal which crystalline phases are present when particles are examined individually. It should not be read as proof that every multicomponent formulation will be completely resolved; overlapping phases, preferred orientation and weak diffraction can still complicate analysis.

MicroED compared with other structure methods
No method wins on every axis. The appropriate choice depends on whether the sample is crystalline, how much material is available, the structural question and the instruments a laboratory can access.
| Approach | Primary evidence | Sample requirement | Typical strength | Important limitation |
|---|---|---|---|---|
| MicroED | Electron diffraction from microcrystals | Suitable nanocrystals or microcrystals on a grid | Direct crystal structures from very small particles; rapid diffraction acquisition in the 2018 examples | Requires crystallinity, electron-microscopy access and specialist diffraction analysis |
| Single-crystal X-ray diffraction | X-ray diffraction from a single crystal | Usually a larger, well-formed crystal | Highly established atomic structures and broad crystallographic workflows | Crystal growth can be the bottleneck for powders or reluctant crystallizers |
| NMR spectroscopy | Nuclear-spin signals in solution or solid state | Appropriate concentration and an NMR-compatible sample | Connectivity, dynamics, purity information and solution-state behavior | Structure is inferred from multiple resonances and can be difficult for mixtures or overlapping signals |
| Mass spectrometry and vibrational spectroscopy | Mass, fragmentation or spectroscopic signatures | Ionizable or spectroscopically responsive material | Composition, functional groups and rapid screening | Usually does not provide a complete three-dimensional crystal structure alone |
For a compound that forms only nanocrystals, MicroED may be the shortest route to a crystallographic answer. For a noncrystalline compound, NMR or other spectroscopies may be more informative. A robust identification often combines orthogonal methods instead of treating one result as a complete chemical answer.
Practical requirements and failure points
Crystallinity and particle selection
The method starts with a physical requirement: at least some particles must have a sufficiently ordered lattice. Amorphous solids, heavily disordered precipitates and poorly crystalline formulations may yield diffuse scattering rather than indexable spots. Screening several particles can reveal whether the problem is sample-wide or limited to a particular crystallite.
Radiation damage and vacuum compatibility
Electrons can damage organic matter. Cryogenic operation, low-dose practices and efficient data collection help, but beam sensitivity remains compound-specific. A particle that diffracts initially can lose order during exposure, so operators must monitor intensity and avoid treating a single damaged pattern as representative.

Mixtures and polymorphs
Powders may contain multiple polymorphs, hydrates, solvates or excipients. Selecting one particle can produce a valid structure for one phase while missing another. Phase identification therefore benefits from examining multiple crystallites and comparing results with complementary bulk measurements.
Instrument and expertise access
MicroED requires a transmission electron microscope configured for diffraction, cryogenic handling, suitable detectors and personnel who understand both microscopy and crystallography. A facility may also impose requirements for grid preparation, sample quantity, hazardous-material handling and data transfer. Performance figures from a published study should not be assumed to match every microscope or service laboratory.
Why the newer organic-solvent cryo-TEM work is a separate development
A 2026 study by Unabara and co-authors, indexed in Microscopy, addressed a different problem: imaging materials in a frozen organic solvent and mapping elements with cryogenic electron energy-loss spectroscopy (cryo-EELS). The researchers formed thin methanol films, froze them into an amorphous state and detected elemental signals from the solvent. They also clearly mapped silicon from mesoporous silica nanoparticles dispersed in methanol.

This is not the 2018 MicroED small-molecule crystallography experiment. MicroED uses diffraction from a crystalline analyte to solve a structure; the methanol work uses cryo-TEM imaging and EELS to study solvent-containing materials and elemental distributions. Both use cryogenic electron microscopy hardware, but their measurements, specimens and scientific outputs differ.

Gradient blotting and vitrified methanol
In a June 11, 2026 account, Tohoku University described a preparation called “gradient blotting.” Only part of the TEM grid contacts filter paper, creating a thickness gradient rather than removing liquid uniformly. The reported method produced regions approximately 100–300 nm thick, vitrified methanol with liquid ethane, and found frozen-methanol beam tolerance comparable to vitrified water under the reported conditions.
The thickness gradient gives operators a better chance of finding an electron-transparent region on the same grid. It is a preparation strategy for this solvent system, not a general recipe that automatically works for every organic liquid.
Can cryo-TEM work with organic solvents?
Yes, but “can” must be qualified by solvent and experiment. Organic liquids can be difficult to vitrify, and some are unstable under an electron beam. Vitrification depends on cooling rate, film thickness, thermal contact and the solvent’s physical properties. Beam exposure can cause radiolysis, bubbling, chemical transformation or loss of imaging quality.

That work establishes why solvent choice matters; it does not make all organic solvents routine cryo-TEM specimens. The 2026 methanol protocol is a specific advance demonstrated for methanol and the reported nanoparticle system.
| Question | MicroED (2018) | Organic-solvent cryo-TEM/EELS (2026) |
|---|---|---|
| What is measured? | Electron diffraction from microcrystals | Images and electron-energy-loss elemental maps in frozen solvent |
| What is the main output? | A crystallographic structure model | Visual structure and elemental distribution in a solvent-containing specimen |
| Key sample condition | Analyte must form suitable microcrystals | Solvent film must be thin, vitrified and sufficiently beam-stable |
| Demonstrated material | Small-molecule powders and other tested crystalline samples | Frozen methanol and mesoporous silica nanoparticles dispersed in methanol |
| Main uncertainty | Whether a particular sample yields an indexable, damage-resistant crystal | Whether a particular solvent can be vitrified and withstand the planned exposure |
How to choose the right electron-based approach
- Define the output. Choose MicroED when you need a crystal structure; choose cryo-TEM/EELS when you need images or elemental distributions in a frozen solvent-containing material.
- Check the physical state. Ask whether the analyte is crystalline and whether suitable particles are likely. For solvent studies, evaluate vitrification and beam stability for that specific liquid.
- Separate screening from proof. A fast diffraction screen can identify promising particles, while NMR, chromatography or spectroscopy may still be needed for purity, concentration, impurities or solution behavior.
- Consult the facility before preparing samples. Grid type, blotting, cryogen, dose limits and transfer procedures are instrument- and laboratory-specific. Follow the facility’s validated protocol rather than copying a published parameter in isolation.
- Validate the interpretation. Inspect diffraction quality, phase assignment, refinement statistics and beam-dose effects. For EELS, confirm that elemental signals are consistent with controls and the specimen geometry.
Bottom line for researchers
MicroED made small-molecule crystallography practical for some nanocrystalline powders: the 2018 study reported approximately 100 nm crystals, sub-1 Å structures and minute-scale data collection in its tested cases. Its “certainty” is the certainty of an unambiguous diffraction-based structure when the sample and experiment succeed, not a universal promise or a substitute for every analytical method.
The 2026 methanol work extends cryogenic electron microscopy in another direction by showing frozen-organic-solvent imaging and cryo-EELS elemental mapping, including silicon in methanol-dispersed silica nanoparticles. It demonstrates what is possible for methanol under a defined preparation, while the broader solvent literature shows that vitrification and beam tolerance remain solvent-specific engineering problems.
