The most practical way to measure galactose oxidase (GalOx) is an initial-rate, horseradish-peroxidase (HRP)-coupled ABTS assay. GalOx oxidizes galactose or a galactoside at the C6 position, producing one equivalent of hydrogen peroxide (H2O2) for each substrate molecule oxidized. HRP uses that peroxide to convert ABTS into a colored radical, which is followed spectrophotometrically near 414–420 nm. Convert the initial absorbance slope with an H2O2 calibration curve, then report activity using an explicitly stated unit definition.
This guide gives a published 96-well implementation, a cuvette-style Fusarium protocol, calculation formulas, controls, troubleshooting, and two alternatives: Amplex Red/HRP for fluorescence-compatible detection and HPAEC-PAD for direct measurement of the oxidized carbohydrate product.
What the assay measures
GalOx is a copper-dependent oxidase that transfers electrons from galactose or a galactoside to molecular oxygen. The carbohydrate is oxidized at C6 to an aldehyde, while oxygen is reduced to hydrogen peroxide. For Fgr GalOx, the stoichiometric description is: oxidation of 1 mole of substrate consumes 1 equivalent of O2 and generates 1 equivalent of H2O2 (Royal Society of Chemistry, 2022).
What’s actually slowing this PC down?
Pick the symptom - the matching free tool is one click away.
In the standard coupled assay, the peroxide is not measured directly. HRP uses it to oxidize 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid), or ABTS, forming a colored radical cation. The increase in absorbance during the initial linear part of the trace is proportional to the peroxide-generation rate, provided HRP and ABTS are not limiting and the blank has been accounted for.

That distinction matters when comparing results. ABTS is a peroxide proxy, whereas direct carbohydrate analysis measures the GalOx product itself. Different publications also use different definitions of a “unit,” so a numerical activity value is meaningful only with its assay conditions and unit convention.
Choose the readout before setting up the experiment
| Method | What is detected | Equipment | Best use | Main limitation |
|---|---|---|---|---|
| HRP-coupled ABTS | H2O2 proxy through colored ABTS oxidation | Visible-range plate reader or spectrophotometer | Routine activity screening and initial-rate measurements | Reporter oxidation, peroxide background, and coupling-enzyme effects can influence the signal |
| Amplex Red/HRP | H2O2 converted to fluorescent or colored resorufin | Fluorescence-capable reader, or a spectrophotometer | Low-background fluorescence workflows and plate assays | Still an indirect peroxide assay and requires suitable fluorescence controls |
| HPAEC-PAD | Oxidized galactosyl product directly | High-performance anion-exchange chromatograph with pulsed amperometric detection | Mechanistic studies, product-resolved kinetics, and enzyme-variant comparisons | Lower throughput and a need for carbohydrate-analysis expertise |
Use ABTS when you need a robust, accessible screen. Consider Amplex Red when fluorescence improves sensitivity or fits the available reader. Use HPAEC-PAD when peroxide coupling could obscure the chemistry or when product identity and formation must be resolved directly.
Published 96-well ABTS/HRP protocol
The following setup is the microplate example reported for Fgr GalOx by the Royal Society of Chemistry in 2022. Treat it as a starting condition, not a universal recipe: pH, substrate concentration, temperature, enzyme loading, wavelength, and unit definitions vary among preparations.
Reagents and equipment
- Transparent 96-well microplate suitable for absorbance measurements.
- 50 mM sodium phosphate buffer, pH 7.0.
- ABTS at 0.25 mg mL−1.
- HRP at 0.1 mg mL−1.
- Galactose or lactose at a selected concentration between 0.1 and 7 mM.
- Fgr GalOx at 5 nM unless the experiment specifies another concentration.
- Hydrogen-peroxide standards spanning 1–10 μM.
- A temperature-controlled plate reader set to 23 °C and capable of reading at 414 nm.
Prepare the plate
- Prepare the phosphate buffer and verify its pH at the temperature used for the assay.
- Make the ABTS/HRP premix in buffer. Keep the composition identical between sample wells, blanks, and standards.
- Add the premix to the wells, reserving positions for a complete reaction, an enzyme-free blank, a substrate-free blank, and the H2O2 calibration series.
- Add GalOx to the reaction wells at the intended final concentration. Add the same volume of enzyme storage buffer to the enzyme-free blank so that volumes and matrix are matched.
- Start each reaction by adding galactose or lactose to the final selected concentration. Add buffer instead of substrate to the substrate-free blank.
- Bring every well to a 100 μL final volume. Mix consistently without creating bubbles.
- Read absorbance at 414 nm at intervals short enough to resolve the initial linear region. Keep the plate at 23 °C during the measurement.
Starting all wells in the same order and using a multichannel pipette or a carefully timed dispensing sequence is important. The slope is time-dependent, so inconsistent delays between substrate addition and the first read can create artificial differences between replicates.
Run the peroxide calibration
Prepare H2O2 standards covering 1–10 μM in the same buffer and reporter matrix used for samples. Read them at 414 nm and fit absorbance to peroxide concentration over the range that is demonstrably linear. Use the resulting slope to convert sample absorbance rates into μM H2O2 per minute. A standard curve made in a different matrix can misrepresent the response if ABTS, HRP, buffer, or plate effects differ.
Check initial-rate linearity
Plot absorbance against time for every reaction and blank. Use only the early interval in which the complete reaction is linear and the blank behaves predictably. If the trace curves immediately, dilute the enzyme or shorten the read window rather than fitting a long interval that includes substrate depletion, reporter limitation, or product effects.
Tube or cuvette implementation
A published Fusarium oxysporum characterization used a different but related format. GalOx was activated with 1 mM CuSO4 for 30 minutes at 25 °C. The assay contained 1 μmol ABTS, 2 U HRP, and 100 μmol galactose in 20 mM potassium phosphate buffer at pH 7.0. Absorbance was followed at 420 nm at 30 °C for 180 seconds.

That paper defined one unit as the amount of enzyme that oxidizes 2 μmol of ABTS per minute, equivalent in its stated convention to consuming 1 μmol of O2 per minute. This is not interchangeable without conversion with the RSC convention of 1 μmol H2O2 produced per minute. Report the source convention alongside any value obtained with this format.
A second Fusarium protocol used the same ABTS/HRP principle at 30 °C and examined substrates including d-galactose and lactose. The differences in buffer concentration, temperature, substrate amount, wavelength, and activation step illustrate why assay conditions must accompany an activity number.
Copper activation: when to include it
Several recombinant GalOx protocols include a copper-loading step of 1 mM CuSO4 for 30 minutes at 25 °C. Copper should not be added automatically to every sample: match the treatment to the enzyme’s preparation history and to the protocol you are reproducing.
If the enzyme may have lost its copper cofactor, measure activity before and after the documented CuSO4 activation step. Include a buffer-treated control so that any change can be attributed to copper rather than to incubation, dilution, or temperature. Excess free copper can also alter sample chemistry, so keep the activation and post-activation dilution consistent across all conditions.
Calculate GalOx activity from an absorbance trace
1. Determine the initial slopes
Fit a straight line to the validated initial region of each complete reaction and its matching blank. Express both slopes as absorbance units per minute:
net slope = slopecomplete reaction − slopeblank
Use the enzyme-free blank for background from ABTS, HRP, substrate, peroxide contamination, and other non-enzymatic processes. The substrate-free blank is useful for identifying reporter oxidation or enzyme-matrix effects that occur without galactose or lactose.
2. Convert absorbance rate to peroxide rate
From the H2O2 standard curve, obtain the calibration relationship between absorbance and peroxide concentration. If the fitted standard-curve slope is m absorbance units per μM H2O2, then:

For volumetric activity:
U mL−1 = μmol H2O2 min−1 ÷ mL enzyme solution added
Under the RSC convention, one unit is 1 μmol H2O2 produced per minute. Under the 2014 Fusarium convention, one unit is defined through oxidation of 2 μmol ABTS per minute. Do not compare values from those papers as though they use the same unit.

Worked calculation template
Suppose a 100 μL well gives a blank-corrected slope of 0.060 min−1, and the peroxide calibration slope is 0.012 absorbance units per μM. The peroxide rate is:
0.060 ÷ 0.012 = 5 μM min−1
The amount rate in the well is:
5 μM min−1 × 0.100 mL ÷ 1000 = 0.0005 μmol min−1
That value becomes a mass- or volume-specific activity only after dividing by the actual GalOx amount. The numbers above illustrate the calculation format; they are not a universal activity result.
Controls that make the result interpretable
- Complete reaction: GalOx, substrate, ABTS, HRP, and buffer.
- Enzyme-free blank: all components except GalOx; detects non-enzymatic signal.
- Substrate-free blank: GalOx, ABTS, HRP, and buffer without galactose or lactose; detects activity or reporter changes unrelated to substrate turnover.
- Peroxide standards: known H2O2 concentrations in the assay matrix; converts optical signal to chemical rate.
- Replicate wells: reveal pipetting variation and allow a mean and dispersion estimate.
- Copper-treatment control: compares untreated and CuSO4-activated enzyme when cofactor occupancy is uncertain.
A rising enzyme-free blank must be subtracted before calculating activity. If the blank is large relative to the complete reaction, identify and correct the source rather than relying on a mathematically corrected but unstable result.
Independent reader supportYour contribution helps us test, update, and keep practical guides available for everyone.Troubleshoot abnormal traces
The blank increases over time
Possible causes include non-enzymatic ABTS oxidation, contaminated or degraded peroxide, substrate or reagent background, and handling contamination. Prepare fresh reagents where appropriate, verify the water and buffer, inspect each component in isolation, and subtract the matched blank only after confirming that its behavior is stable and reproducible.
The reaction trace is curved from the first read
The selected interval may already include substrate depletion, reporter limitation, mixing artifacts, or an enzyme concentration outside the linear range. Lower the GalOx concentration, shorten the read window, improve mixing and timing, or reduce the substrate and reporter loads only after confirming that the assay remains sensitive.

Check that substrate was actually added, that HRP and ABTS are present at their intended final concentrations, and that the reader is using the correct wavelength. If the enzyme preparation may be apo- or partially copper-depleted, compare it with the documented CuSO4-activation treatment. Also verify that the first read was not delayed long enough for a short initial-rate phase to be missed.

Replicates disagree
Look for bubbles, edge-well temperature differences, inconsistent dispensing order, incomplete mixing, and unequal enzyme incubation. Randomize or balance the order of additions, avoid bubbles, and use identical timing for every well.
ABTS appears unsuitable for the sample
ABTS methods can be affected by peroxide cross-reactivity and instability of the ABTS radical reporter. If matrix components produce background or distort the reporter response, move to a fluorescence-compatible Amplex Red/HRP assay or measure the oxidized carbohydrate directly by HPAEC-PAD.
Amplex Red/HRP as a fluorescence-compatible alternative
In an Amplex Red assay, GalOx-generated H2O2 is converted by HRP and Amplex Red into resorufin. The product can be measured fluorometrically or spectrophotometrically. The A22179 manual lists approximate resorufin maxima of 571 nm for absorbance and 585 nm for fluorescence (Thermo Fisher Scientific, manual; publication year not stated on the accessed page).
Recommended Free Tools
Use the same experimental logic as ABTS: include a complete reaction, enzyme-free and substrate-free controls, a peroxide or resorufin calibration appropriate to the chosen readout, and an initial-rate analysis. Fluorescence is not automatically free of interference; assess sample autofluorescence, quenching, and reagent background in the actual matrix.
Direct product measurement with HPAEC-PAD
High-performance anion-exchange chromatography with pulsed amperometric detection (HPAEC-PAD) quantifies the oxidized galactosyl product rather than inferring turnover from peroxide. This is useful when the peroxide reporter is unstable, when sample components react with ABTS or Amplex Red, or when you need to distinguish products during mechanistic or enzyme-variant studies.
The trade-off is practical: HPAEC-PAD requires chromatography hardware, carbohydrate-analysis expertise, sample preparation, calibration standards, and substantially lower throughput than a plate reader. It is therefore better suited to product-resolved kinetic work than to rapid screening.
How to report a GalOx activity measurement
A reproducible result should state all variables that can change the rate:
